内皮素和一氧化氮对大鼠急性胰腺炎病变进展的影响

摘    要:目的:探讨血浆内皮素(ET)和一氧化氮(NO)水平变化在大鼠急性胰腺炎病变进展中的作用。方法:42只SD大鼠分急性胰腺炎组和对照组,进行不同处理。急性胰腺炎组用十二指肠闭襻法制作急性胰腺炎模型,对照组仅作胃窦切开缝合术,在病程不同时点测定血浆淀粉酶、ET、NO水平,并观察胰腺组织病理学改变。结果:急性胰腺炎组随着病变进展,血浆淀粉酶升高,病程12h内,ET与NO同步上升,ET与NO比值维持相对平衡,胰腺炎病理主要表现为水肿性;病程12h后ET继续升高,而NO则升高不明显,ET与NO比值升高,病程24h胰腺炎病理主要表现为出血坏死性,胰腺组织病理学评分显著升高。对照组病程中各指标均呈低水平变化。结论:急性胰腺炎病程中,ET产生增多,而NO产生不足,ET与NO平衡失调,是急性胰腺炎病变进展的主要因素之一。

Abstract:Objective: To explore the effects of endothelin(ET) and nitric oxide(NO) on the progression of acute pancreatitis(AP) in rats.Methods: 42 SD rats were randomized into two groups,AP and control group.The SD rats in AP group were all made into AP model through the closed duodenal loop technique;however,the SD rats in control group were merely performed sinus ventriculi discussion and suturation.Then the contents of plasma amylase(AMS),ET and NO were measured respectively,and the pathological changes of pancreatic tissue were observed.Results: In AP group,accompany with the progression of acute pancreatitis in SD rats,the level of AMS were increased,and within 12 hours,the levels of ET and NO were increased synchronously,and the pathological changes were edematous pancreatitis;after 12 hours,the level of ET was increased continuously,however,the level of NO was not increased significantly,the ratio between ET and NO was increased,and the pathological changes were hemorrhagic and necrotic pancreatitis.In control group,the levels of AMS,ET and NO had few changes.Conclusion: The increased ET generation and insufficient NO generation result in the imbalance between ET and NO,and that might play a key role in the development of AP.
