摘 要: | 目的 研究特异性微小RNA(miR)-590对舌鳞癌细胞SCC-3 Fas配体(FasL)的表达的影响.方法 将合成的miR-590 mimics或miR-590抑制剂转染至SCC-3细胞.通过RT-PCR和Western blot法检测SCC-3中FasL信使RNA(mRNA)和蛋白的表达;将转染后的SCC-3细胞在顺铂(DDP)5 mg/L处理下,在不同时间段通过MTT法绘制细胞生长曲线,并通过针对FasL的miR-590的抑制剂从反面验证miR-590的作用.结果 miR-590可以在mRNA和蛋白水平上降低SCC-3细胞FasL的表达;转染miR-590后SCC-3细胞对于DDP的耐药性降低.结论 miR-590可以有效抑制SCC-3细胞FasL的表达,降低SCC-3的DDP的耐药性,从而可能降低和减少其对免疫活性细胞的凋亡诱导和杀伤作用,从而阻断肿瘤细胞的免疫逃逸,为舌鳞癌的生物治疗提供了新的靶点.

Abstract: | Objective To investigate the effect of miR-590 on the expression of FasL in SCC-3 cells.Methods The synthetic miR-590 mimics or miR-590 inhibitor was transfected into SCC-3 cells.The mRNA and protein expression of FasL mRNA and protein in SCC-3 were detected by RT-PCR and Western blot respecitively.The transfected SCC-3 cells were treated with Cisplatin(DDP) 5 mg/L and the cell growth curves were drawn at different time points by MTT assay, and the effect of miR-590 was verified from the negative side by the inhibitor of miR-590 against FasL.Results MiR-590 decreased the expression of FasL on SCC-3 cells at mRNA and protein levels;Resistance to DDP was decreased in SCC-3 cells after miR-590 transfection.Conclusion MiR-590 can effectively inhibit the expression of FasL in SCC-3 cells and decrease the resistance of SCC-3 to DDP, which may reduce the apoptosis-inducing and killing effect of immunocompetent cells, blocking tumor immune escape,providing a new target for the treatment of tongue squamous cell carcinoma. |
