摘 要: | 目的 探讨microRNA-210(miR-210)对乳腺癌细胞增殖、侵袭和凋亡的影响.方法 采用RNA干扰技术,构建针对miR-210的shRNA真核表达载体,转染MCF-7细胞,采用实时荧光定量PCR技术检测转染后miR-210的表达情况,观察转染后MCF-7细胞的增殖、侵袭和凋亡情况.结果转染后MCF-7细胞中miR-210表达明显下降,同时细胞增殖和侵袭能力减弱、凋亡率上升.结论 miR-210能够影响乳腺癌的生物学行为.

Abstract: | Objective To explore the effect of microRNA-210 (miR-210) on proliferation, invasion and apoptosis of breast cancer cell. Methods The plasmid expression vector of short hairpin RNA ( shRNA) targeting miR-210 was constructed and transfected into MCF-7 cell by LipofectamineTM2000. The expression of miR-210 was detected by Real-time fluorescence quantitative PCR method after transaction, and proliferation, invasion, apoptosis of MCF-7 cell were observed. Results The expression of miR-210 decreased significantly in MCF-7 cell, and prolif-erative and invasive ability weakened, and cell apoptosis rate increased after transfection with miR-210 shRNA. Conclusion miR-210 expression can affect biological behavior of breast cancer. |
