摘    要:天然高分子聚合物聚L-谷氨酸(PLGA)与氧化海藻酸钠(OAg)共价交联合成PLGA/OAg水凝胶,将人脂肪来源干细胞(hADSCs)接种于水凝胶内,倒置显微镜、扫描电镜观察细胞在支架中的存活、增殖、黏附情况,DNA定量法检测细胞的增殖。体外证实该水凝胶材料生物相容性良好,具有性能可控性和微创性等优势,对于进一步开发脂肪组织工程材料具有指导性意义。

Biocompatibility research of hADSCs cultivated in PLGA/OAg hydrogel in vitro

Abstract:PLGA/OAg hydrogel was fabricated by poly L-glutamic acid(PLGA) and oxidation sodium alginate(OAg) covalent crosslinking.HADSCs were encapsulated into the PLGA/OAg hydrogel scaffolds and cultivated in three-dimensional environment in vitro to observe biocompatibility of the scaffolds include cell viability,adhesion,proliferation by inverted microscope and scanning electron microscopy.The cell proliferation was tested by DNA quantitative method.The synthetical hydrogel performances an excellent biocompatibility,controllability and minimally invasive advantages.It may be an instructive fat tissue engineering material in the future.
