组织工程人工神经对大鼠坐骨神经缺损后神经再生速度的影响

摘    要:目的研究组织工程人工神经对大鼠坐骨神经缺损后神经再生速度的影响。方法建立15mm的SD大鼠右侧坐骨神经缺损模型后,用嗅球成鞘细胞(olfactory ensheathing cells,OEC),雪旺氏细胞(Schwann cells,SC),细胞外基质成份(extracellular matrix,ECM)以及自行制备的聚乳酸-聚羟基乙酸共聚物(poly(DL-lactide-coglycolide),PLGA)导管,构成OEC-SC-ECM-PLGA桥接体进行修复,同时设OEC-ECM-PLGA组、SC-ECM-PLGA组、ECM-PLGA组、PLGA组和自体神经移植组进行对照。在术后1,3,6,9周行神经再生速度检测。结果术后1、3、6w,由于新生神经纤维无法着色而无法计算神经再生速度,术后9w,OEC-SC-ECM-PLGA组和自体神经移植组的神经再生速度均为(0.71±0.00)mm/d,显著快于OEC-ECM-PLGA组的(0.60±0.05)mm/d和SC-ECM-PLGA组(0.60±0.09)mm/d(P〈0.05)。ECM-PLGA组和PLGA组神经再生速度均为零。结论 OEC-SC-ECM-PLGA桥接体可显著提高大鼠坐骨神经缺损后神经再生速度。

Effect of tissue-engineering artificial nerve on the neuro-regeneration speed after rat sciatic nerve defect

Abstract:Objective To explore the effect of tissue-engineering artificial nerve on neuro-regeneration speed after rat sciatic nerve defect.Methods The 15mm SD rat right sciatic nerve defect model was established and repaired with OEC-SC-ECM-PLGA bridging complex,which contained OEC,SC,ECM,and self-made PLGA conduit.At the same time,OEC-ECM-PLGA group,SC-ECM-PLGA group,ECM-PLGA group,PLGA group,and nerve autograft group were also set up as the controls.At 1th week,3th week,6th week,9th week after surgery,the neuro-regeneration speed test were performed.Results From 1 weeks to 6 weeks after surgery,it is impossible to calculate the neuro-regeneration speed.From 6 weeks to 9 weeks after surgery,the neuro-regeneration speed of OEC-SC-ECM-PLGA group and nerve autograft group were(0.71±0.00)mm/d,which were faster than that of OEC-ECM-PLGA and SC-ECM-PLGA groups(P0.05).Conclusions OEC-SC-ECM-PLGA bridging complex can significantly enhance the neuro-regeneration speed after rat sciatic nerve defect.
