茶多酚对雷公藤甲素致小鼠胃粘膜损伤的保护作用

摘    要:目的:观察不同剂量雷公藤甲素(Tri)致小鼠胃粘膜损伤的作用以及茶多酚(TP)对其损害的保护作用,并初步探讨相关机制。方法:昆明种小鼠70只,雌雄各半,随机分成5组:高剂量Tri组(0.03 mg/kg)、低剂量Tri组(0.015mg/kg)、高剂量Tri(0.03 mg/kg)+TP(300 mg/kg)组、低剂量Tri(0.015 mg/kg)+TP(300 mg/kg)组和正常对照组。各组按相应剂量每日灌胃给药60 d后观察各组小鼠胃粘膜损伤指数,测定胃组织匀浆MDA含量、SOD活性并行胃粘膜病理组织学检查。结果:与对照组相比,低剂量Tri组、高剂量Tri组均可见胃粘膜明显损伤,胃粘膜损伤指数显著升高(P<0.05),胃组织匀浆MDA的含量显著提高,SOD活性显著降低(P<0.05),病理组织学均可见显著胃粘膜上皮细胞变性坏死等改变;高剂量Tri+TP组、低剂量Tri+TP组胃组织匀浆MDA含量、SOD活性无显著性改变,病理组织学轻度异常。结论:Tri对小鼠胃粘膜有一定的损害,而TP具有很好的保护作用,其机制与对抗组织脂质过氧化反应有关。

Abstract:Objective:To study the effects of tea polyphenols(TP) on protecting gastric mucosa damage of mice induced by triptolide(Tri)and explore the correlated mechanism.Methods: Seventy mice were randomly divided even into five groups: high dose Tri(0.03 mg/kg) group,low dose Tri(0.015 mg/kg)group,high dose Tri(0.03 mg/kg)+ TP(300 mg/kg)group,low dose Tri(0.03 mg/kg)+ TP(300 mg/kg) group and control group.Each group was given Tri of the above dosage via mouth per day for sixty days for observing gastric damage index,superoxide dismutase(SOD) activities and malondialdehyde(MDA) contents in gastric tissue homogenate and the pathohistology of gastric tissue.Results: The activities of SOD and content of MDA between high dose Tri + TP group,low dose Tri + TP group and control group did not show remarkable difference.However,in the high dose Tri group and low dose Tri group,the contents of MDA were higher((P<0.05))and the activities of SOD were lower than the control group(P<0.05).The pathohistologic changes such as cell degeneration and cellular necrosis of gastric mucosa epithelium in the high dose Tri group and low dose Tri group were notably observed,while the pathohistology in the other three groups were not significant.Conclusion:WTBZ] Tri may result in gastric mucosa damage of mice while TP can decrease the damage efficiently.The mechanism is relevant to reducing lipid peroxidation.
