PTEN和VEGF的表达与宫颈鳞癌血管生成关系的研究

摘    要:目的:研究宫颈鳞癌组织中抑癌基因(PTEN)和血管内皮生长因子(VEGF)的表达与宫颈癌血管生成的关系。方法:应用免疫组织化学S-P法检测45例宫颈鳞癌组织和20例慢性宫颈炎组织(对照组)中PTEN和VEGF的表达,同时分析不同PTEN和VEGF表达状态下,宫颈癌组织微血管密度(MVD)的变化。结果:宫颈癌组和对照组PTEN阳性率分别为51.1%(23/45)和95.0%(19/20),2组比较差异有统计学意义(P<0.01);宫颈癌组和对照组VEGF阳性率分别为60.0%(27/45)和30.0%(6/20),2组比较差异有统计学意义(P<0.05);PTEN和VEGF的表达呈负相关(rs=-0.595,P<0.01)。同时根据PTEN和VEGF的表达状况将宫颈癌分为4组,组1为PTEN阴性VEGF阳性(n=20),组2为PTEN阴性VEGF阴性(n=2),组3为PTEN阳性VEGF阳性(n=7),组4为PTEN阳性VEGF阴性(n=16)。4组平均MVD分别为31.13、18.17、20.90、21.94,组1显著高于其他3组(P<0.05)。结论:宫颈癌PTEN基因失活可能通过增加VEGF的表达来促进血管生成,导致肿瘤恶性进展。

Abstract:Objective:To study the relationship of expression of PTEN and vascular endothelial growth factor(VEGF)with angiogenesis in cervical squamous carcinoma. Methods:The expression of PTEN and VEGF from 45 cases with cervical squamous carcinoma and 20 cases with normal cervical tissues(control group) was detected with immunohistochemical S-P method for examining variation of PTEN and VEGF in cervical squamous carcinoma tissues and microvessel density (MVD). Results:The positive rates of PTEN in cervical squamous carcinoma and control groups were 51.1% (23/45) vs 95.0% (19/20) and 60.0%(27/45) vs 30.0% (6/20) in VEGF, respectively, which was significantly different between the 2 groups(P<0.01). PTEN and VEGF expression showed negative correlation (r-s=-0.595, P<0.01). When the group of cervical squamous carcinoma were subdivided into groups of PTEN-negative and VEGF-positive cases (group A , n=20), PTEN-negative and VEGF-negative cases(group B, n=2), PTEN-positive and VEGF-positive cases(group C, n=7) or PTEN-positive and VEGF-negative cases (group D, n=16), the mean MVD of groups A, B, C and D was 31.13, 18.17, 20.90 and 21.94, respectively.The mean MVD of group A was significantly higher than that of 3 other groups(P<0.05). Conclusion:Inactivation of PTEN gene in cervical squamous carcinoma may promote angiogenesis through pathway of increase in VEGF expression, thus inducing tumor progression.
