热疗联合放疗对S180细胞凋亡及血管再生的影响

摘    要:目的 探讨热疗联合放疗对小鼠S180肿瘤模型细胞凋亡及血管再生的影响.方法 实验用昆明小鼠60只,右后肢股部皮下接种S180肿瘤细胞株,当肿瘤生长至1cm3时分组进行局部热疗、放疗和热放疗.然后用RTPCR检测Bcl-2 mRNA,免疫组化方法检测调亡细胞,血管内皮生长因子(VEGF)蛋白,以微血管密度(MVD)作为定量检测指标.结果 热疗、放疗、热放疗组细胞凋亡指数较对照组增加(P<0.01),以热放疗组增加最为显著(P<0.01).热疗、放疗、热放疗组Bcl-2基因表达较对照组减弱(P<0.01),以热放疗组减弱最为显著(P<0.01).热疗组VEGF蛋白表达和MVD较对照组降低(P<0.01),放疗组VEGF蛋白表达和MVD较对照组升高(P<0.01),热放疗组VEGF蛋白表达和MVD较放疗组降低(P<0.01).结论 热疗可以下调Bcl-2基因,诱导S180株细胞凋亡,抑制肿瘤细胞VEGF的产生,使MVD下降.与放疗共同作用于肿瘤细胞,可增加肿瘤细胞放射敏感性,对肿瘤有显著的治疗作用.

Abstract:Objective To study the effects of hyperthermia combined radiation on apoptosis and angiogenesis of S180 tumor cells.Methods S180 tumor cells were injected subcutaneously into 60 rats in back-right leg,which were randomly assigned into 4 groups.When the tumors volume reached 1cm3,hyperthermia,radiation,hyperthermia combined radiation were given respectively.Then Bcl-2 mRNA was measured by RT-PCR;Apoptosis,VEGF and MVD were measured by immunohistochemical staining.Results Compared with the control group,the apoptosis fraction of hyperthermia(H),radiation(R),hyperthermia combined radiation(HR) group were increased(P<0.01),the Bcl-2 mRNA were decrease(P<0.01),the VEGF and MVD of H were decreased(P<0.01),that of R were increased(P<0.01).The VEGF and MVD of HR were decreased compared R(P<0.01).Conclusion Hyperthermia can depress Bcl-2 mRNA,induce S180 cell apoptosis,and restrain tumor cells produce VEGF protein.Hyperthermia combined radiation can improve the tumor cell sensitivity to irradation and become a better therapy to tumor patients.
