甘肃省半夏种质资源遗传多样性分析

摘    要:运用荧光标记引物的扩增片段长度多态性(AFLP)分子标记技术,对50份甘肃半夏样品进行了DNA指纹图谱构建和遗传差异分析。选出的8对AFLP引物共扩增出517条带,其中396条具有多态性,多态性位点百分率为75.8%,平均每对引物扩增出64.6条带和49.5个多态性条带。系统聚类分析结果表明所有半夏AFLP指纹均不相同,来源地相同的半夏表现出相对密切的亲缘关系,甘肃省西和县的半夏同天水市的半夏遗传差异明显。聚类分析结果获得了Bootstrap校验的支持。甘肃省半夏遗传多样性十分丰富,AFLP分子标记可有效应用于半夏种质资源鉴别和遗传多样性分析。

Abstract:The DNA fingerprint and genetic variation of 50 individuals of Pinellia ternata (Thunb.) Breit from Gansu Province were analyzed by fluorescence labeled AFLP technique. A total of 517 DNA bands and 396 polymorphic DNA bands were detected by eight primer pairs screened from 64 primer pairs. The percentage of polymorphic bands was 75.8 %, total amplified bands and polymorphic bands per primer combination produced were 64.6 and 49.5, respectively, indicating a high level of genetic diversity existed in P. ternata in Gansu Province. The phylo-genetic cluster analysis revealed that all the individuals showed independent DNA fingerprints and could be absolutely differentiated in our assay. Individuals from similar regions were mainly clustered together and exhibited closer genetic relationship. Meanwhile, individuals from region of Longnan showed clear genetic distinction from those of Tianshui. The clustering results were strongly supported by Bootstrap testing. Our findings indicated that the genetic diversity of P. ternata was abundant in Gansu Province. AFLP markers can be potentially used as an effective tool for studying the genetic diversity and germplasm identification of P. ternata.
