枸杞多糖对2型糖尿病大鼠血糖、血脂及TNF-α水平的影响

摘    要:目的观察枸杞多糖对2型糖尿病大鼠血糖、胰岛素、血脂及血清TNF-α水平的影响。方法采用高脂高糖饲料喂养8周联合腹腔注射小剂量链脲佐菌素的方法,建立2型糖尿病大鼠模型。将未予上述处理的大鼠设为正常对照组,成模大鼠按血糖再随机分为糖尿病对照组、枸杞多糖(LBP)低、中、高剂量干预组和二甲双胍组,药物干预6周后,处死动物取血测定血脂、空腹血糖,应用放射免疫分析法测定空腹胰岛素及血清TNF-α水平。结果LBP中、高剂量干预组及二甲双胍组空腹血糖水平较糖尿病对照组降低(P〈0.05,P〈0.01),较正常对照组高(P〈0.05);LBP高剂量干预组及二甲双胍组ISI较糖尿病对照组升高(P〈0.05);LBP中、高剂量干预组血清TC水平较糖尿病对照组降低(P〈0.05),LBP各剂量组血清TNF-α水平较糖尿病对照组有所下降,但差异无统计学意义(P〉0.05)。二甲双胍组血清TNF-α水平较糖尿病对照组降低(P〈0.05)。结论LBP能够降低2型糖尿病大鼠血糖及血脂水平,增加组织的胰岛素敏感性,改善糖尿病大鼠糖脂代谢紊乱及胰岛素抵抗。

Abstract:Objective To study the effect of Lycium Barharum Polysaccharides( LBP ) on blood glucose, serum insulin, blood lipid and Tumor necrosis factor - α( TNF - α) in type 2 diabetic rats. Methods The model of type 2 diabetic rats was induced by high - fat and high - sugar diet feeding for 8 weeks combined with intravenous injection of a small dose of streptozotocin( STZ). Then the ruts without above - mentioned treatment were selected as the normal contral group. The diabetic rats were randomly divided into five groups: the model group and low, middle and high doses of LBP groups and metformin group. The course of treatment lasted for 6 weeks. Then all ruts were killed, and their blood was extracted for measuring the levels of blood lipid and fasting blood glucose. The levels of fasting insulin and serum TNF - α were tested by rudioimmunoassay. Results The level of fasting blood glucose in the middle and high doses of LBP group and mefformin group was lower than that in the model group(P 〈0.05 ,P 〈0. 01 ), and was higher than that in normal control group(P 〈 0, 05 ). Insulin sensitive index ( ISI ) in the high dose of LBP group and mefformin group was higher than that in the model group( P 〈0.05). The level of serum total cholesterol in the middle and high doses of LBP group was lower than that in the model group (P 〈0.05). The level of serum TNF - α in different doses of LBP group was lower than that in the model group, but without statistical sense (P 〉 0. 05 ). The level of serum TNF - α in the mefformin group was lower than that in the model group (P 〈 0.05). Conclusion LBP can decrease the levels of blood glucose and blood lipid of type 2 diabetic ruts, increase insulin sensitivity of the target tissues, improve metabolism disorder of glucose and lipid, and ameliorate insulin resistance of diabetic rats.
