摘    要:目的:观察高浓度葡萄糖对人血管内皮细胞周期和凋亡的影响。方法:用不同浓度葡萄糖(Glu)作用体外培养的人脐静脉内皮细胞(HUVECS)24h、48h、72h,绘制细胞增殖曲线,用流式细胞仪分析细胞周期和凋亡的变化。结果:高浓度Glu(20mmol/L、40mmol/L)对内皮细胞的生长增殖有明显抑制作用,使G0/G1期细胞比例增加,S和G2/M期细胞比例下降,同时诱导了内皮细胞的凋亡,并随作用浓度增加、时间延长更为明显。结论:高糖使培养的HUVECS凋亡加速,同时抑制细胞增殖。细胞可能被阻滞在G1/S转变期。

Abstract:Objective:To investigate the effects of high glucose on cell cycle and apoptosis in the cultured human umbilical vein endothelial cells (HUVECS). Methods: HUVECS were treated with different concentrations of glucose (5,20,40 mmol/L) for 24,48,72 hours.The cell proliferation curve was made to observed the inhibition effects of glucose on HUVECS growth.Cell cycle and apoptosis were measured by flow cytometry. Results: High-glucose(20,40 mmol/L) significiently inhibited HUVECS proliferation(P<0.01) and accelerated cell apoptosis in a dose and time dependent manner than the low-glucose(5mmol/L) group(P<0.01).In the meanwhile ,the percent of the cultured cells in G 0/G 1 phase in high glucose were increased, the percent of S and G 2/M phase were decreased (P<0.05) .Conclusion: High-glucose(20mmol/L?40mmol/L) may inhibit the cell proliferation and accelerated cell apoptosis of HUVECS. Endothelial cells were possibly arrested in G 1/S checkpoint.
