摘    要:目的探索外源性硫化氢对THP-1源性巨噬细胞脂质蓄积的影响及机制。方法以硫氢化钠(NaHS)做为外源性硫化氢的供体,采用油红O染色结合光密度法测细胞内脂质含量,RT-PCR和Westernblot法测SR-A mRNA和蛋白表达。结果氧化型低密度脂蛋白(ox-LDL)显著诱导巨噬细胞内脂质蓄积和SR-A表达,而NaHS呈浓度依赖性降低ox-LDL诱导的细胞内脂质蓄积,并显著下调SR-A mRNA和蛋白水平。结论外源性硫化氢可能通过抑制SR-A表达减少ox-LDL诱导的THP-1源性巨噬细胞脂质蓄积。

Abstract:Objective To explore the effects and mechanisms of sodium hydrosulfide(NaHS,an exogenous H2S donor) on lipid accumulation in THP-1-derived macrophages. Methods Oil O staining,RT-PCR and western blot were performed.
