结核杆菌异烟肼耐药性产生的分子机制研究

摘    要:本文设计了两对引物,分别采用PCR扩增结核杆菌异烟肼耐药性相关基因—KatG基因酶活性中心区,并用单链构象多态性分析(SSCP)及DNA序列分析方法对临床分离的9株异烟肼敏感株、11株异烟肼中、低度耐药株进行了耐药性分子机制的研究,结果发现中国人结核杆菌产生异烟肼耐药性的主要原因是由于katG基因发生碱基点突变所致,本资料亦为快速检测结核杆菌异烟肼耐药性研究方法提供资料。

Abstract:wo pairs of primers were designed.Nine isolates of isoniazid-sensitive and 11 isolates ofisoniazid-resistant M.tuberculosis were analyzed by polymerase chain reaction amplification of396bp sequence of katG gene.With the use of single-strand conformation polymorphism analysisand DNA sequencing,8 isolates in 396bp products and 5 isolates in 2l6bp products of the ll iso-lates showed point mutations and no deletion or insertion was detected.Thus,in China,pointmutations in katG gene can mainly account for inactivation of catalase-peroxidase,leading to iso-niazid resistance.The results provide laboratory evidence for detecting the isoniazid resistance in M.tuberculosis and also explore its molecular mechanism.
