磷酸钙骨水泥/顺铂复合体体外药物缓释及体内抑瘤和修复骨缺损的实验研究

摘    要:目的:寻找磷酸钙骨水泥/顺铂复合体修复肿瘤性骨缺损的最佳质量构成比。方法:通过原子吸收分光光度法对通过混合-模压法制备的不同质量比构成的磷酸钙骨水泥/顺铂复合体体外缓释液中顺铂浓度进行测定,寻找最合适的磷酸钙骨水泥、顺铂质量构成比,在此基础上制备骨缺损和移植肿瘤动物模型和动物体内进行骨缺损的修复实验和体内抑瘤实验。结果:含0.1%~0.2%顺铂的磷酸钙骨水泥能够修复兔骨缺损,同时又能对肿瘤细胞产生抑制杀伤作用。结论:含0.1%~0.2%顺铂的磷酸钙骨水泥能够消灭残存肿瘤细胞,且最终不影响骨修复的进程。

Abstract:Objective To explore the best mass ratio of calcium phosphate cement (CPC) / cisplatin complex filling and to repair bone defect caused by tumor resection. Methods Mixed-molding method was used to obtain cisplatin/calcium phosphate cement complex at 0, 0.1%, 0.2%, and 0.4% mass ratio. Drug concentration was determined by atomic absorption spectrophotometry. Bone defect of rabbits and osteosarcoma of rats were prepared. We implanted CPC and CPC/cisplatin complex to observe the repair of bone defect and the inhibition of tumor in vivo. Results CPC containing 0.1% ~ 0. 2 % cisplatin not only repaired the bone defect in rabbits but also eliminated osteosarcoma in rats. Conclusion CPC containing 0.1% ~ 0. 2 % cisplatin can repair bone defect and eliminate tumor without influencing the prosthetic precess.
