摘    要:目的 探讨过氧亚硝基阴离子(ONOO^-)在脂多糖(LPS)所致急性肺损伤中的作用。方法 用免疫组织化学技术检测内毒素休克时ONOO^-生成的标志物硝基酷氨酸在大鼠肺脏内的定位与分布。将不同剂量的外源性ONOO^-经气管快速推注入健康大鼠肺脏,定量检测肺系数、肺湿干重比、肺水含量和伊文思蓝含量,光镜观察单层蓝标记肺微血管数量及病理变化。结果 LPS静脉注射2h和6h后,肺脏有明显损伤及硝基酷氨酸产

Abstract:Objective Toexploretherolesofperoxynitrite(ONOO-)inacutelunginjuryinducedby lipopolysaccharides (LPS) Methods Nitrotyrosine (NT) ,aspecificmarkerofinvivoONOO-production ,was detectedwithimmunohistochemicaltechniqueinratlungduringendotoxinshock (LPS 5mg/kgiv) Lung coefficient(LC) ,lungwet/dryweightratio (W/D) ,thecontentsofwaterandEvansblue(EB)inlungswere measuredafterintratrachealinsufflationofONOO-withdifferentdoses Monastralblue (MB) labelledblood vesselsandpathologicalchangesinlungswereexaminedunderalightmicroscope Results After 2or 6hoursof LPSintravenousadministration ,severelunginjuryandpositivesignalsofNToccurredwiththelatterlocatedinthe pulmonarymacrophages,endothelia ,subendothelialandmuscularlayersofthepulmonaryartery Administrationof exogenousONOO-causeddose dependentincreaseinLC ,W /D ,waterandEBcontentsandinthenumberofMB labelledbloodvesselsinratlungs,togetherwithsignificantpathologicalalterationssuchasdiffusealveolar collapse ,capillarycongestionintheseptaandfocalhemorrhage Conclusion Intheearlystageofendotoxin shock,severelunginjuryandONOO-productionsimultaneouslyexist,andexogenousONOO-couldinduce significantdysfunctionofmicrovascularendothelialbarrierandlunginjury,indicatingthatONOO-generationin vivomaybeinvolvedinthepathogenesisofacutelunginjury
