摘    要:为阐明癌症发生的内在机制和探索肿瘤分化治疗与基因治疗的合适途径,作者应用10^-5mol/L全反式维甲酸诱导人肺腺癌细胞系GLC-82,观察到(1)细胞体外生长速度减慢,堆叠性生长消失,细胞出出现空泡,细胞逐渐衰老和死亡;(2)双层软琼脂中细胞集落形成爱到明显抑制;(3)细胞DNA合成能力降低;(4)细胞周期出现向G1/G0期移行的特征性动力学改变;(5)细胞分化和凋亡的效应器因子-谷氨酰胺转移酶

Abstract:All-trans-retinoic acid(RA)is a powerful differentiation-inducing reagent.We examined the effect of RA on malignant phenotype of human lung adenocarcinoma cell line GLC-82 in vitro.Treatment of GLC-82 cell with 10-5mol/L.RA for 1-7 days resulted in suppression of cell proliferation(33-55%),inhibition of colony formation in soft agar(97.5%),and a decrease of3H-TdR incorporation(30-60%).Cytokinetic studies demonstrated that the cells arrested in G1/G0 phase increased from 36.0% to 72.4%,which is typical for cell differentiation.Human endothelial cell transglytaminase(TGase)was expressed persistently during RA treaament.Treatment of GLC-82 cell with RA gave rise to senescence and apoptosis gradually.The results indicated that induction of differentiation and modulation of gene expression can be achieved by RA treatment in human lung adenocarcinoma cell line GLC-82.
