MSP法检测胰腺癌Syk基因甲基化改变的研究

目的 :探讨胰腺癌Syk基因启动子区 5’CpG岛甲基化改变的特点与临床病理特征的关系。方法 :采用甲基化特异性PCR(MSP)方法检测Syk基因启动子甲基化情况。结果 :32例癌旁正常胰腺组织未发现有Syk基因启动子的甲基化 ,14 /32例胰腺癌组织检测到Syk基因启动子的甲基化 ,癌组织Syk基因启动子甲基化率显著增高 (P <0 .0 5 )。有淋巴结转移的 15例胰腺癌组织中 ,有 10例Syk基因启动子甲基化 ,有淋巴结转移的Syk基因启动子甲基化显著高于无淋巴结转移组 (P <0 .0 5 )。结论 :Syk基因启动子甲基化是导致Syk基因失活的原因之一 ,Syk基因启动子的甲基化与胰腺癌的发生、转移相关。

Detection of methylation status of Syk gene in pancreatic carcinoma by MSP

Objective: To detect methylation status of the 5'CpG island locating in the promoter region of Syk gene in pancreatic carcinoma, the relationship between the aberrant methylation of Syk gene and clinical manifestation was analyzed. Methods:Using methylation-specific PCR technique(MSP), specimens from 32 pancreatic carcinoma patients(tumor tissues, adjacent normal tissues)to detect methylation of Syk promoter region. Results:No methylation of Syk promoter was found in adjacent normal tissues, 14 cases in 32 pancreatic carcinoma patients(43.7%) were detected hypermethylation of Syk gene in promoter region, the rate of methylation of Syk promoter in pancreatic carcinoma was higher than that of in adjacent normal tissues(P<0.05). In 15 cases with lymph node metastasis, 10 were found with Syk promoter methylation. A significant difference was noted between two groups(P<0.05). Conclusion:Hypermethylation leads to silencing of the Syk gene in human pancreatic carcinoma. Methylation of Syk promoter is correlated to oncogenesis, metastasis of pancreatic carcinoma.
