摘    要:目的:探讨胸苷酸合成酶基因多态性与肿瘤发生易感性的关系。方法:通过检索PubMed, EBSCO和 OVID等数据库,纳入37个关于胸苷酸合成酶基因多态性和肿瘤易感性相关性的病例对照研究。采用RevMan 5.0统计软件进行文献的异质性检验并计算合并效应优势比(OR)和95%可信区间(CI)。结果:经筛选纳入37个病例-对照研究,其中包含13307例肿瘤病例和18826个正常对照, 胸苷酸合成酶5`UTR 28bp处串联重复序列和3`UTR 6bp处ins/del(insertion/deletion)变异 2个位点的OR和CI分别是(3R/3R versus 2R/2R:OR=1.06,95%CI,0.93-1.20)、(del6/del6 versus ins6/ins6:OR=0.93,95%CI,0.81-1.08),对肿瘤类型和人群进行亚族分析时也未发现明显关联性。结论: 胸苷酸合成酶基因5`UTR 28bp处串联重复序列和3`UTR 6bp处ins/del两个位点多态性与肿瘤发生不存在明显关联性。

Thymidylate Synthase Genetic Polymorphisms and Cancer Risk:A Meta-analysis of 37 Case-Control Studies
Abstract:Objective:To evaluate the association between genetic variants in Thymidylate Synthase(TS) and cancer risk using a meta-analysis. Methods:A comprehensive search was conducted to identify all case-control studies on TS on a 28bp tandem repeats in 5`Untranslated Region (5`UTR) and a 6 bp insertion(ins) and deletion(del) mutation in 3`UTR of the gene and cancer risk. Meta-analysis was conducted using a fixed and random effect model. Results:Our meta-analysis on a total of 13307 cancer cases and 18226 control subjects from 37 published case-control studies showed no significant association between the risk of cancer and the 5`UTR 28bp tandem repeats polymorphism(3R/3R versus 2R/2R:OR=1.06,95%CI,0.93-1.20) or the 3`UTR 6 bp ins/del polymorphism(del6/del6 versus ins6/ins6:OR=0.93,95%CI,0.81-1.08) with significant between-study heterogeneity. In the cancer type- and ethnic subgroup-stratification analyses, we didn`t find any association between TS polymorphisms and cancer risk either. Conclusion:The analyses suggest that TS 5`UTR 28 bp tandem repeats and 3`UTR 6 bp ins/del polymorphisms may not be associated with cancer risk.
