Platelet-derived growth factor of astrocyte expression and functional recovery in diabetic spinal cord injury rats

Abstract:Background Prospective mortality study revealed that mortality was elevated in diabetes compared to US rates in chronic spinal cord injury (SCI). Our study was conducted to investigate the platelet-derived growth factor (PDGF) of astrocyte expression changes in spinal cord injury on streptozotocin (STZ)-induced diabetic rats. Methods Thirty male SD rats were randomly divided into 3 groups: spinal cord injury group, diabetic spinal cord injury group and sham operation control group. We employed streptozotocin (STZ)-induced diabetic Sprague-Dawley rats for four weeks and used a weight-drop contusion SCI model. Rats were sacrificed on day 7 after induction of spinal cord injury model. The immunohistochemistry and western blot analysis of were used to detect the PDGF expression level. BBB was also used to evaluate the neurological recovery level of the rats. Results 7 days after acute spinal injury, the diabetic spinal injury group showed the slower recovery speed in motor function as lower BBB score. We also found the PDGF positive astrocytes number and light density were significantly more high and intensive in SCI group than the diabetic SCI group (P<0.05). Conclusion PDGF as an important factor for the recovery of neurological function after acute spinal injury and the hyperglycemia in diabetic rats could depress the expression of PDGF in injured spinal cord, which may help to explain the slower recovery and higher mortality in diabetes after spinal cord injury.
