摘    要:目的以聚氰基丙烯酸丁酯纳米粒(PBCA-NPs,NPs)作为端粒酶逆转录酶(hTERT)反义寡核苷酸(ASODN)载体转染肺腺癌A549细胞,观察其对该细胞hTERT mRNA表达的影响。方法以NPs携带ASODN转染A549细胞后,用流式细胞仪检测细胞周期;逆转录-聚合酶链反应(RT-PCR)检测细胞hTERT mRNA表达的改变。结果转染后,与空白对照组和正义寡核苷酸(SODN)组相比,ASODN组G0/G1期细胞增加,S期细胞明显减少(P<0.01),hTERT mRNA表达明显下降(P<0.01)。结论hTERT ASODN能有效改变A549细胞的细胞周期,下调hTERTmRNA表达,对肺腺癌细胞生长有抑制作用。

Abstract:Objective To investigate the effect of hTERT antisense oligodeoxynucleotide on the gene expression of hTERT mRNA by polybutylcyanoacrylate nanoparticles(PBCA-NPs,NPs).Methods After transfecting hTERT ASODN by NPs into A549 cell,cell cycles were analyzed by flow cytometry(FCM),the expression of hTERT mRNA was determined by RT-PCR.Results Compared with control group and(SODN-)NP group,the cell cycles in ASODN-NP group varied obviously and the cell cycle was blocked in G_0/G_1 phase obviously(P<0.01),the expression of hTERT mRNA of ASODN group decreased observably((P<)0.01).Conclusion hTERT ASODN mediated by PBCA-NPs can vary A549 cell cycles and reduce the expression of hTERT mRNA,it can inhibit the growth of A549 cell efficiently.
