血红素氧合酶-一氧化碳系统在肠易激综合征中的表达

摘    要:目的检测血红素氧合酶-一氧化碳(HO—CO)系统在肠易激综合征(IBS)患者中的表达,以探讨其在肠易激综合征发病机制中的作用。方法①采用比色法测定各组IBS患者和对照组血浆中CO的浓度。②采用免疫组织化学SABC染色法观察HO-1、HO-2在IBS患者及对照组的乙状结肠黏膜中的定位;并用计算机图像分析技术半定量监测其蛋白表达的强弱,平均光密度越高说明表达越强。结果①腹泻型IBS患者血浆中CO的浓度明显高于对照组和便秘组(P〈0.05);②HO-2和HO-1都主要在结肠黏膜上皮细胞中表达,其中HO-2在IBS组和对照组的表达差异无统计学意义,而HO-1在腹泻组IBS患者中的表达明显强于对照组和便秘组(P〈0.05)。结论HO—1在IBS患者结肠中的表达异常以及CO的浓度在IBS患者血浆中的异常改变,表明HO—CO系统在IBS等胃肠动力性疾病的发病机制中可能起重要作用。

Abstract:Objective To detect the change of HO - CO system in the patient with irritable bowel syndrome was investigated. Its effect on the pathogenesis of irritable bowel syndrome were discussed. Methods The distribution of HO - 1 and HO - 2 proteins in the colonic tissues in 38 cases of IBS and 15 cases of normal control group were detected by SABC immunohistochemistry and perfusion catheter manometer. The slides were then analyzed with an image analyzing system to obtain the density(mean) of stained proteins. The concentrations of CO in the plasma were determined with chemical methods. Results HO - 1 and HO - 2 were expressed mainly in the colonic mucous membrane epithelia. The expression of HO - 1 in patients with diarrhea predominant IBS was higher than that in normal group and constipation IBS. The con- centrations of CO in diarrhea predominant IBS was significantly higher than that in the normal control group and constipation IBS. Conclusions The abnormal expression of HO - CO system in IBS suggests that CO may plays an important role in the pathogenesis of gastrointestinal motility disorders such as IBS.
