葛根素对2型糖尿病大鼠脂代谢及脂肪分化相关蛋白基因表达的影响

摘    要:目的观察葛根素(Pue)对2型糖尿病(T2DM)大鼠脂代谢及脂肪分化相关蛋白(ADRP)基因mRNA表达的影响。方法大鼠高糖高脂饮食伴尾静脉注射小剂量链脲佐菌素(STZ)25mg/kg复制T2DM大鼠模型,将造模大鼠随机分为模型组(腹腔注射丙二醇)和3个Pue治疗组(腹腔注射40、80、160mg/kg的Pue),另选10只大鼠为正常对照组(腹腔注射丙二醇)。持续腹腔注射6周后,测定各组大鼠空腹血糖(FBG)、血清胰岛素(FINS)和血脂水平,并计算胰岛素敏感指数(ISI);同时以RT-PCR法检测脂肪组织ADRP基因mRNA水平。结果模型组大鼠FBG、FINS、总胆固醇(TC)、三酰甘油(TG)、高密度脂蛋白-胆固醇(HDL-C)、低密度脂蛋白-胆固醇(LDL-C)、游离脂肪酸(FFA)水平及ISI与对照组比较,差别有统计学意义(P<0.05);Pue高、中剂量组大鼠FBG、FINS、TC、TG、LDL-C、FFA水平及ISI与模型组比较,差别有统计学意义(P<0.05),Pue低剂量组大鼠FBG、TC、TG、FFA水平及ISA与模型组比较,差别有统计学意义(P<0.05)。与对照组比较,模型组大鼠脂肪组织ADRP基因mRNA表达显著增高(P<0.01),Pue高、中剂量组大鼠ADRP基因mRNA表达较模型组下降(P<0.05)。结论Pue可通过降低T2DM大鼠脂肪组织ADRP基因mRNA表达,改善糖脂代谢紊乱和胰岛素抵抗。

Abstract:Objective To observe the effects of puerarin(Pue)on lipid metabolism and adipose differentiated-related protein(ADRP)gene mRNA expression in rats with type 2 diabetes mellitus(T2DM).Methods Rats were fed with high glucose and fat diet and injected with small dose of streptozocin(STZ)25 mg/kg to set up T2DM model.The model rats were randomly divided as model control group and three Pue groups(40,80,160 mg/kg)with 10 in each group,another 10 rats were selected as normal control group.Six weeks later,the levels of fasting blood glucose(FBG),fasting insulin(FINS)and blood lipid were measured,and insulin sensitivity index(ISI)was calculated.,ADRP gene mRNA in fatty tissue was determined by RT-PCR.Results Compared with model control group,the differences in levels of FBG,FINS,total cholesterol(TC),high density lipoprotein-cholesterol(HDL-C),lower density lipoprotein-cholesterol(LDL-C)free fatty ac id(FFA)and ISI in the model group,had a statistic significance(P<0.05);the differences in levels of FBG,FINS,TC,TG,LDL-C,FFA and ISI in the high and middle dosage of puerarin groups had a statistic significance(P<0.05);the differences in levels of FBG,TC,TG,FFA and ISI in the lower dosage of puerarin group had a statistic significance(P<0.05);Compared with model control group,the ADRP gene mRNA expression of fatty tissue in the model group obviously increased(P<0.01),but the expressions in the high and middle dosage of puerarin groups decreased obviously(P<0.05).Conclusion Puerarin could improve lipid metabolism disorders and insulin resistance by decreasing the ADRP gene mRNA expression in T2DM.
