摘    要:目的:探讨JNK对胰岛素和葡萄糖刺激下脂肪细胞表达脂联素(adiponectin)和抵抗素(resistin)的影响。方法:用不同浓度葡萄糖(5mmol/L,25mmol/L)和胰岛素(0.1nmol/L,100nmol/L)刺激分化成熟的脂肪细胞,各浓度均设立JNK抑制剂组(SP600125),采用荧光定量PCR检测adiponectin和resistin的mRNA表达。结果:高浓度胰岛素和葡萄糖均能使adiponectinmRNA表达降低(P<0.01),resistin表达升高(P<0.01),对应的抑制剂组adiponectin表达较高浓度组增加(P<0.05),resistin表达降低(P<0.05);两个低浓度组与对应抑制剂组及对照组相比均无明显差异。结论:高浓度葡萄糖和胰岛素影响了脂肪细胞脂联素和抵抗素的表达,JNK在其中起到了介导作用。

Abstract:Objective: To investigate the effects of JNK on insulin-or glulcose-mediated adiponectin and resistin mRNA expression in 3T3L1 adipocytes. Methods: Different concentrations of glucose(5 mmol/L,25 mmol/L) and insulin(0.1 nmol/L,100 nmol/L) were added to the culture mediator for 3T3L1 adipocytes pretreated with or without JNK inhibitor SP600125. The mRNA expression of adiponectin and resistin were evaluated by quantitative fluorescence PCR. Results: Adiponectin expression was significantly decreased by high levels of insulin or glucose,whereas the resistin expression increased. Furthermore, both increased adiponectin and decreased resistin expression were blocked by pretreatment with SP600125 in high level groups. There was no significant difference in adiponectin and resistin expression among groups treated with low levels of insulin or glucose with or without SP600125. Conclusion: JNK is involved in high levels of insulin-or glucose-mediated adiponectin and resistin mRNA expression in 3T3L1 adipocytes.
