细胞色素P450 2E1基因多态性与食管癌易感性的关系

目的:研究CYP2E1基因多态性和烟酒嗜好的相互作用,以及其与食管癌易感性的关系。方法:采用病例-对照分子流行病学方法,以聚合酶链反应-限制性片段长度多态性(PCR-RFLP)方法分析78例食管癌患者和118例正常对照者的CYP2E1基因型。结果:①食管癌组吸烟、饮酒状况分布与对照组之间差异有显著性(P值均<0.01),而CYP2E1基因Rsa I位点和Dra I位点各基因型频率在食管癌组和对照组的分布差异无显著性(P值分别为0.141、0.376)。②Loglistic回归分析表明,CYP2E1基因Rsa I位点和吸烟、饮酒都是食管癌的危险因素,其优势比(OR)分别为1.5、3.1、6.4,但是CYP2E1基因的C1/CI基因型和DD基因型与吸烟、饮酒之间未发现存在协同致癌性。结论:CYP2E1 C1/C1基因型和吸烟、饮酒都是食管癌的危险因素,而且食管癌组吸烟、饮酒状况分布与对照组之间差异有显著性。

Impact of CYP2E1 Polymorphisms on the Risk of Esophageal Cancer

To evaluate the relationship among polymorphism with CYP2E1 gene, habit of smoking and alcohol, and their interaction on the risk of esophageal cancer. Methods: Genotyping of CYP2E1 was performed using PCR-RFLP analysis in 78 esophageal cancer cases and 118 normal matched controls. Results: A significant difference was observed in the habit of smoking and alcohol between controls and esophageal cancer cases, but no significant difference of CYP2E1 genetypes was observed. By multivariate analysis, the CYP2E1 C1 /C1 genetype, smoking and alcohol are the risk factors of esophageal cancer, and the odds ratio(OR) are 1.5, 3.1 and 6. 4 respectively. Smoking and alcohol are important risk factors of esophageal cancer, but they can not interact with CYP2E1 C1/C1 genetype and CYP2E1 DD genetype to increase the risk of esophageal cancer. Conclusion:The CYP2E1 C1/C1 genetype and smoking and alcohol are important risk factors for development of esophageal cancer, but there is no an apparent gene - environment interaction found between them.
