休克/再灌注肝组织中氧自由基的直接测定及其意义

摘    要:运用自旋捕捉技术和ESR直接测定兔休克/再灌注肝组织中氧自由基的变化,并进行肝组织光镜观察。实验结果表明:休克1.5小时后,肝内氧自由基有明显增高,再灌注15分钟、30分钟后有更明显的增高,在再灌注前运用CAT治疗,能使氧自由基显著降低,但仍有很高水平。肝病理改变光镜表现为休克1.5小时肝细胞浊肿,再灌注后细胞呈点状及片状坏死,而运用CAT治疗能使之改善。提示氧自由基在休克/再灌注肝组织损伤起了重要作用。

Abstract:Electron spin resonance spectroscopy and spin trapping technique were used to directly measure oxygen free radical of rabbit liver tissue following shock/reperfu- sion.The experimental results showed that 1.5 hours after shok oxygen free radical of the liver tissueed increased significangly with for further increase following reperfusion 15 min- utes and 30 minutes.Oxygen free radical was decreased by treatment with catalase(CAT) before reperfusion, but the level was still high.Under light microscope,pathologic changes of the liver tissue such as cloudy swelling of hepatocytes might be found at the end of 1.5 hours of shock,individual liver cell necrosis and focal hepatocellular necrosis might be seen after reperfusion.These findings suggest that oxygen free radical may play an important role in shock/reperfusion injury of the liver.
