神经生长因子对缺氧缺血性脑损伤细胞凋亡的影响

摘    要:目的:探讨新生儿缺氧缺血性脑损伤(HIBD)与凋亡之间的关系,研究神经生长因子(NGF)对HIBD的保护作用,为用NGF治疗新生儿HIBD提供理论依据。方法:生后7天SD大鼠40只制成HIBD动物模型,随机分成两组:HIBD模型对照组20只,HIBD模型NGF治疗组20只,观察NGF对HIBD模型新生大鼠体重增长情况、死亡率、左右脑重量的影响,并用原位缺口末梢标记(TUNEL)法检测NGF对缺氧缺血后脑细胞凋亡的影响。结果:HIBD模型NGF治疗组体重增长明显高于对照组(P<0.01);治疗组实验过程中死亡率明显低于对照组:HIBD模型NGF治疗组与对照组健侧(右侧)脑重量相近,但治疗组患侧(左侧)脑重量明显重于对照组(P<0.01);治疗组左右脑重量无显著差异,而对照组左右脑重量差异显著(P<0.01);TUNEL染色所见阳性凋亡细胞,其凋亡特征为胞体缩小、核固缩、核碎裂、凋亡小体形成。NGF治疗组阳性细胞均数明显低于对照组(P<0.01)。结论:NGF治疗减轻了缺氧缺血后脑萎缩和细胞凋亡。

Abstract:Objective:To explore the relationship between hypoxic ischemic brain damage (HIBD) and neuronal apoptosis, and evaluate protective eff ect of nerve growth factor (NGF) on apoptosis in hypoxicischemic encephalopathy of newborn rats. Methods:The HIBD models were perpared on 7 day old SD rats, These rats were randomly divided into NGF treated and control groups. Body weights, mortalities and brain weights of NGF treated group were compared with these of control group. The neuronal apoptosis was observed by terminal deo xynucleotidyl transferase mediated dUTP biotin nick end labeling (TUNEL) staini ng. Results:The body weights in NGF treated group were significantly higher than those in control group ( P<0.01 );The death rate in NGF treated group was much lower than that in control group; The right brain weights were s imilar in two groups, but the left brain weights in NGF treated group was significant ly higher than that in control group ( P< 0.01); The average sum of positive cells of TUNEL staining in NGF treated group was significantly lower than that in control group ( P<0.01 ). Conclusion:NGF could reduce neuronal a poptosis and brain damage after hypoxic ischemic insult.
