El-Tor生物型霍乱弧菌可溶性血凝素的纯化

用改进的Finkelstein的方法从Wg2(El-Tor 生物型)弧菌菌株中提纯的可溶性血凝素有下列特点:(1)纯化的血凝素(SHA)具有凝集红细胞的作用,且对不同来源的红细胞的敏感性不同。(2)纯化的SHA具有蛋白酶的水解活性。(3)经免疫双扩散、免疫电泳检测,纯化的SHA,粗制SHA与纯SHA抗血清反应均呈单一沉淀线。与霍乱肠毒素无反应。(4)聚丙烯酰胺凝胶电泳,SDS-凝胶电泳的结果与文献报道基本一致。

PURIFICATION OF THE SOLUBLE HEMAGGLUTININ FROM AN EL-TOR VIBRIO CHOLERAE

Vibrio cholera produces a soluble hemagglutinin(SHA).This protein, a polymer of 32000 Mr subunits,appears to participate in the attachment of vibrio cholera onto the intestinal epithelium.It also displays protease activity.Specific antibody against the purified SHA inhibits not only its protease activity but also the attachment of V-cholerae on the intentinal epithelium.This protein may be also responsible for the cleavage of the subunit A of cholera enterotoxin resulting in its activation.(1)This pro- tein was tested for SHAa activity on chicken erythrocytes(RBC).(2)pro- tease activity was determined,using bovine serum albumin as the sub- str are,on polyacrylamide gel electrophoresis.(3)The purified SHA gives a single precipitation line in immunodiffusion with rabbit anti-SHA se- rum raised agasnst it,but not with antiserum against cholera toxin.(4) By means of SDS-polyacrylamide gel electrophoresis,it y(?)elded two fragments(Mr=32000,Mr=17000).
