丹参对肝硬化门脉高压大鼠缺血再灌注胃粘膜损伤防治作用的研究

摘    要:制成肝硬化门脉高压(PHT)大鼠胃粘膜缺血再灌注模型,用电子自旋共振技术测定缺血再灌注前后及丹参治疗后胃粘膜中氧自由基(OFR)以及脂质过氧化物(LPO)和超氧化物的歧化酶(SOD)的变化,同期观察其光镜、电镜病理改变,探讨丹参对胃粘膜损伤的保护作用。结果表明:PHT大鼠胃粘膜在缺血再灌注过程中有大量OFR产生,粘膜损伤严重程度与OFR、LPO及SOD密切相关;PHT胃粘膜更易受缺血再灌注损伤;早期使用丹参可显著降低胃粘膜中OFR及LPO含量,提高SOD活性,减轻胃粘膜的损伤,起到保护胃粘膜的作用

Abstract:The purpose of the present study is to evaluate the therapeutic effects of Radix Salviate Miltorrhizae(RSM) on gastirc mucosa in ischemia reperfusion with portal hypertension(PHT) rats. PHT models were made in SD rats by injecting CCL 4. The changes of oxygen free radicals(OFR) were measured directly by electron spin resonance(ESR) spectroscopy, with concomitant monitoring amount of lipid peroxide (LPO), activity of superoxide dismutase(SOD) and pathology of gastirc mucosa. Results of the study revealed that the amount of OFR was increased significantly( P <0.01) after ischemia reperfusion in case of PHT in comparison with that of the control group. Gastric mucosa in PHT was more susceptible to ischemia reperfusion insult than normal controls. The pathological changes were in accord with atteration of OFR,LPO, SOD. After RSM treatment levels of OFR and LPO dropped down sharply ( P <0.01), activity of SOD was significantly( P <0.01) higher than that in PHT ischemia reperfusion group.RSM when used at early stage served to protect the gastric mucosa.
