Bcl-2、Bax基因表达改变与禁食诱导的脂肪细胞凋亡研究

摘    要:目的:研究禁食对大鼠体重及外周脂肪湿重的影响,探讨禁食减脂减重与脂肪细胞凋亡的关系。方法:禁食48h,用DNA提取物凝胶电泳证实脂肪细胞凋亡的存在,逆转录(RT)-PCR探讨Bcl-2与Bax基因的表达。结果:(1)禁食能明显降低大鼠体重,组间比较具有显著的统计学意义(P<0.001)(2)禁食能减少外周脂肪组织湿重,但统计学意义不显著(P>0.05);(3)DNA提取物凝胶电泳结果证实脂肪细胞发生了凋亡;(4)外周脂肪组织中Bcl-2基因表达减少,Bax表达增加(5)生殖器周脂肪组织对细胞凋亡的敏感性高于腹膜后。结论:降低Bcl-2与Bax的比值,诱导脂肪细胞凋亡可能是禁食减脂减重的重要机制。

Abstract:Objective:To study the influence of fasting on the body and adipose tissue weight of rats and the relation between fasting and white adipocyte apoptosis. Methods:Adipocyte apoptosis and the expression of Bcl 2 and Bax gene were detected by DNA Ladder and RT PCR in male SD rate fasted in 48 hours. Results:Fasting significantly reduced body weight ( P <0.001),but slightly decreased the weight of adipose tissue ( P >0.05). Adipocyte apoptosis was clearly observed by ladder pattern.Bcl 2 gene expression decreased while Bax increased in fasted adipose tissue. And the epididymal adipocyte was more sensitive to apoptosis than retroperitoneal adipocyte. Conclusion:The decreased Bcl 2 gene expression and increased Bax gene expression may be an important mechanism of adipocyte apoptosis induced by fasting.
