Bcl-2、Bax基因表达改变与禁食诱导的脂肪细胞凋亡研究

目的：研究禁食对大鼠体重及外周脂肪湿重的影响，探讨禁食减脂减重与脂肪细胞凋亡的关系。方法：禁食48h,用DNA提取物凝胶电泳证实脂肪细胞凋亡的存在，逆转录（RT）－PCR探讨Bcl-2与Bax基因的表达。结果：（１）禁食能明显降低大鼠体重，组间比较具有显著的统计学意义（P＜0．001）（2）禁食能减少外周脂肪组织湿重，但统计学意义不显著（P＞0．05）；（3）DNA提取物凝胶电泳结果证实脂肪细胞发生了凋亡；（4）外周脂肪组织中Bcl-2基因表达减少，Bax表达增加（5）生殖器周脂肪组织对细胞凋亡的敏感性高于腹膜后。结论：降低Bcl-2与Bax的比值，诱导脂肪细胞凋亡可能是禁食减脂减重的重要机制。

Bcl-2 and Bax Are Involved in the White Adipocyte Apoptosis Induced by Fasting

Objective:To study the influence of fasting on the body and adipose tissue weight of rats and the relation between fasting and white adipocyte apoptosis. Methods:Adipocyte apoptosis and the expression of Bcl 2 and Bax gene were detected by DNA Ladder and RT PCR in male SD rate fasted in 48 hours. Results:Fasting significantly reduced body weight ( P <0.001),but slightly decreased the weight of adipose tissue ( P >0.05). Adipocyte apoptosis was clearly observed by ladder pattern.Bcl 2 gene expression decreased while Bax increased in fasted adipose tissue. And the epididymal adipocyte was more sensitive to apoptosis than retroperitoneal adipocyte. Conclusion:The decreased Bcl 2 gene expression and increased Bax gene expression may be an important mechanism of adipocyte apoptosis induced by fasting.
