冠脉内皮细胞在电解性氧自由基损伤离体大鼠心脏中的作用

摘    要:为了探讨冠脉血管内皮细胞在外源性氧自田基致心肌损伤中的作用、本文采用电解法产生氧自由基(OFR).观察OFR对保留冠脉内皮与去冠脉内皮的Langendorff大鼠心脏的心率(HR)、心律、冠脉灌注压(CPP)、左室收缩压(LVSP)、左室舒张未压(LVEDP).冠脉流出液中乳酸脱氢酶(LDH)含量,心肌组织中丙二醛(MDA)及Na^ -K^ ATPase水平等的影响。结果:去冠脉内皮组的CPP、LVEDP、LDH、MDA及心律失常严重指数(ASl)均显著低于保留内皮组.而Na^ -K^ ATPase与HR则明显高于保留内皮组。两组的LVSP无明显差异。表明:冠脉由皮细胞可能介导OFR对心肌的损伤、去冠脉羽皮细胞可减轻OFR对心肌的损伤.同时提示:冠脉内皮细胞在心肌缺血再灌注损伤中发挥重要作用。

Abstract:In order to research the role of endothelial cells of coronary vessels in the oxygen free radical (OFR) caused myocardial damage, we observed those parameters that were heart rate and rhythm, coronary perfusion prassure (CPP), left ventricular systolic pressure (LVSP), left ventricular end - diastolic prerssure (LVEDP), level of lactate dehydrogenase (LDH) in the coronary excretion fluid, content of malonadehyde (MDA) and Na - K ATPase in the myocardial tissue in Langendorff rat hearts, which were perfused with OFR generated by eiectrial stimulation, with and without the endothelial cells in the coronary vessels (ECCV). The results showed that CPP, LVEDP, LDH, MDA and ASI in the group without ECCV were rnarkedlg lower than those in the group with ECCV, respectively, but and Na - K ATPase were obsiously above that in the group with ECCV. The results suggested that ECCV might mediate myocardial damage caused by OFR and removing ECCV could reduce this damage. According to this results , we might deduce that coronary vessel endothelial cells play an important role in the myocardial ischemia - reperfusion injury.
