参丹健胰丸对糖尿病大鼠肾醛糖还原酶活性的影响

摘    要:目的:评价参丹健胰丸对糖尿病大鼠肾醛糖还原酶(aldose reductase,AR)活性的影响,探讨中药防治糖尿病肾脏病变的意义.方法: 用中药参丹健胰丸和醛糖还原酶抑制剂螺旋已内酰脲(sorbinil)对糖尿病Wistar大鼠模型进行治疗,并观察肾组织AR活性、尿蛋白和Na -K -ATP酶活性的变化.结果: 糖尿病鼠模型组其AR活性显著高于正常鼠组(P< 0.01),使用sorbinil、参丹健胰丸后AR活性明显下降(P<0.01),与正常对照组相比较无显著性差异(P >0.05).治疗8周末,两个治疗组尿蛋白水平与4周末相当,但糖尿病模型组尿蛋白明显升高,远高于两个治疗组(P<0.01),治疗后两组间无明显差异(P>0.05).用sorbinil和参丹健胰丸治疗后,Na -K -ATP酶活性显著升高(与模型组相比P<0.05).结论: 参丹健胰丸可通过抑制糖尿病大鼠肾脏组织AR活性而发挥其防治效用.

Abstract:Objective:To evaluate the effects of Shendan J ianyi Capsule (SDJYC) on aldose reductase (AR) activity of the kidney in diabe tic rats and the significance of prevention and treatment of diabetic nephropath y with Chinese natural herbs. Methods: Diabetic Wistar rats were tre ated with SDJYC and sorbinil respectively, and the AR activity in kidney, urine protein and the activity of Na +-K +-ATPase were tested. Results : The AR activity in diabetic rats was decreased obviously after being treated w ith sorbinil or SDJYC. The activity of Na +-K +-ATPase in diabetic rats decr eased obviously at the end of the eighth week, and increased after being treated with sorbinil and SDJYC. There was no difference between two treated groups aft er the treatment. It demonstrated that the inhibition of AR activity could impro ve the activity of Na +-K +-ATPase and SDJYC had the same effect. Conc lusions: SDJYC exerts effects by inhibiting the activity of AR .
