葛根素对实验性大鼠电离辐射损伤的保护效应

摘    要:目的:观察葛根素对实验性大鼠电离辐射损伤的保护效应,探讨其保护作用的可能机制.方法:Wistar大鼠随机分为4组,每组8只:生理盐水非辐射(physiological saline non-radiation,SN)组,葛根素非辐射(puerarin non-radiation,PN)组,生理盐水辐射(physiological saline radiation,SR)组,葛根素辐射(puerarin radiation,PR)组.辐射源为60C0γ源,1次辐射剂量为18 Gy,流量为1.2 Gy/min.葛根素为注射针剂,辐射后连续静脉注射6 d,每次剂量为30 mg/kg.辐射1周后取样测定.结果:葛根素处理能够有效缓解辐射引起的大鼠外周血红细胞和白细胞的下降;减轻辐射大鼠胸腺指数和脾脏指数的下降;提高心肌组织超氧化物歧化酶(superoxide dismutase,SOD)活性和降低膜脂过氧化产物丙二醛(malondialdehyde,MDA)的含量.结论:葛根素对实验性大鼠电离辐射损伤有明显的保护效应,其保护效应的机制可能与葛根素的抗氧化作用有关.

Abstract:Objective: To observe the protective effects of puerarin o n radiation injury of experimental rats and to discuss the possible mechanism of its radiation protection. Methods: Wistar rats were divided rando mly into 4 groups with 8 rats in each group: physiological saline non-radiati on (SN) group, puerarin non-radiation (PN) group, physiological saline radi ation (SR) group, puerarin radiation (PR) group. The source of radiation was cob alt-60 gamma rays, and the rats were exposed to radiation (1.2 Gy/min) at a dos e of 18 Gy. Following irradiation of the rats, puerarin was injected intravenous ly at each dose of 30 mg/kg in 6 consecutive days (24 h interval). Samples w ere collected and assayed one week later. Results: Puerarin dela yed effectively the declines of the quantity of red blood cells and white blood cells in circulation due to ionizing irradiation, improved the index of thymus a nd spleen in ionizing irradiation rats, increased the activity of superoxide dis mutase (SOD) and decreased the level of malondialdehyde (MDA) in the rat myoca rdial tissue. Conclusion: Puerarin has a marked protective effect on ionizing irradiation injury of experimental rats, and its protective mech anism is probably based on oxidation-resistance.
