亚硒酸钠对人结肠癌细胞的抑制机制

摘    要:目的:研究亚硒酸钠对人结肠癌细胞的抑制机制。方法:通过DNA末端原位标记染色法(TUNEL)检测亚硒酸钠溶液对人结肠癌细胞株SW-111C凋亡的影响;应用免疫组织化学染色技术观察亚硒酸钠对人结肠癌细胞株SW-111C p16,p21,PCNA,CD44基因表达的影响;并测定亚硒酸钠对小鼠T淋巴细胞亚群的影响。结果:TUNEL染色法表明亚硒酸钠作用细胞后凋亡指数明显升高(P<0.01),且具有剂量-效应关系;亚硒酸钠作用后人结肠癌细胞SW-111C p16和p21基因的蛋白表达上调,PCNA和CD44基因的蛋白表达下调;亚硒酸钠能使小鼠CD3 ,CD4 细胞数增加,而且CD4 /CD8 比值升高,与对照组比较差异有统计学意义(P<0.05)。结论:亚硒酸钠可通过诱导细胞凋亡,影响p16,p21,PCNA,CD44基因表达和提高机体的免疫功能而抑制SW-111C的生长。

Abstract:Objective:To investigate the inhibition mechanism of sodium selenite on SW-111C cells.Methods: The apoptosis of SW-111C cells treated with sodium selenite was observed by TUNEL.The expression of p16,p21,PCNA,CD44 treated with sodium selenite was performed by immunohistochemistry technique.The T-lymphocyte subsets were detected by animal experiment. Results: The apoptotic index of SW-111C cells increased and was positively correlated to the concentration of sodium selenite(P<0.01).The expression of p16 and p21 was up-regulated and the expression of PCNA and CD44 was down regulated after treated by sodium selenite.After exposure to sodium selenite,the number of CD3 and CD4 cells in mice increased and CD4 /CD8 increased too(P<0.05). Conclusion: The sodium selenite could inhibit the proliferation of SW-111C cells by inducing cell apoptosis,and influence the expression of p16,p21,PCNA,CD44 with improving immune function.
