胰腺癌组织FOXC2的表达及其与预后的关系

目的: 探讨叉头框C2(forkhead box C2，FOXC2)在胰腺癌中的表达及其与胰腺癌患者预后的相关性。方法: 利用Oncomine数据库分析FOXC2 mRNA在胰腺癌及癌旁组织中的表达；收集112例胰腺患者癌组织及相应癌旁组织，采用qRT-PCR和免疫组织化学染色检测FOXC2的表达，并分析其与临床病理参数的关系。应用Kaplan-meier法分析FOXC2表达对患者生存时间的影响。结果： Oncomine数据库中FOXC2 mRNA在胰腺癌癌组织中表达明显高于癌旁组织(t_Badea=4.074，t_Pei= 3.741，P均<0.05)；FOXC2蛋白主要表达于胰腺癌细胞胞核，且癌组织免疫组化评分明显高于癌旁组织( t=7.571，P<0.05)；FOXC2 mRNA在胰腺癌组织中表达明显高于癌旁组织( t=13.79，P<0.05)；有淋巴结转移的癌组织中FOXC2阳性表达率高于无淋巴结转移的癌组织(χ ² =16.998，P<0.01)，胰腺癌Ⅲ期FOXC2阳性表达率高于Ⅰ期( χ 2=15.688，P<0.001)和Ⅱ期( χ ²=10.678，P=0.001)。FOXC2阳性组胰腺癌患者的中位生存时间显著短于阴性组( χ ²=10.32，P=0.001 3)，FOXC2阳性表达(HR=2.569，P=0.003)可作为预测胰腺癌预后的独立因素。 结论： FOXC2在胰腺癌组织中呈高表达，且与胰腺癌患者的预后高度相关。

The relationship between FOXC2 expression and prognosis in pancreatic cancer

Objective: To investigate the expression of forkhead box C2(FOXC2) in pancreatic cancer and its correlation with the prognosis of patients with pancreatic cancer. Methods: Oncomine database was used to analyze the expression of FOXC2 mRNA in pancreatic cancer and adjacent tissues of cancer. Real time PCR and immunohistochemical staining were used to detect the expression of FOXC2 in 112 cases of pancreatic cancer and adjacent tissues. The relationship between FOXC2 expression and the clinical parameters was analyzed. Kaplan-Meier method was used to analyze the association of FOXC2 expression with the survival time of patients. Results: FOXC2 mRNA was up-regulated in pancreatic cancer tissues compared with adjacent tissues by analyzing Oncomine database(t_Badea=4.074,t_Pei=3.741,both P<0.05).The expression level of FOXC2 mRNA in pancreatic cancer tissue was significantly higher than that in adjacent tissues (t=13.79,P<0.05). The positive rate of FOXC2 in cancer tissues with lymph node metastasis was higher than that without lymph node metastasis(χ ²=16.998,P<0.01). The positive rate of FOXC2 in stage Ⅲ was higher than that in stage Ⅰ (χ ²=15.688,P<0.001) and stage Ⅱ (χ ²=10.678,P=0.001). The median survival time of FOXC2 positive group was significantly lower than that of the negative group ( χ ²=10.32,P=0.001 3). The positive expression of FOXC2 (HR=2.569,P=0.003) was an independent predictor for prognosis of pancreatic cancer. Conclusion: FOXC2 expression was up regulated in pancreatic cancer tissue, which was highly related with the prognosis of patients with pancreatic cancer.
