低氧预适应对大鼠移植肝脏功能和组织结构的影响

摘    要:目的:探讨低氧预适应对肝脏移植大鼠术后早期肝脏病理结构和肝功能改变的影响.方法:用"二袖套法"建立大鼠原位肝脏移植模型, A组:原位肝移植组,B组:低氧预适应组,C组:正常大鼠对照组.对3组大鼠进行肝功能测定及常规病理观察,统计一周生存率.结果:术后B组与A组相比较肝功能损害明显减轻、组织结构得到更好的保存、一周生存率显著提高(P<0.05).结论:低氧预适应可能通过提高肝细胞耐受肝移植时较严重的缺氧损伤,维持细胞的能量状态,减轻移植肝脏的病理损害,改善移植术后的存活率.

Abstract:Objective: To investigate the effect of the hypoxic precondition(HP) on rats liver earlier-stage pathology and function changes after transplantation.Methods: Rats orthotopic liver transplantation(OLT) were according to Kamada two-sleeve approach.Adult male Sprague-Dawley(SD) rats were randomly divided into three groups,A: OLT group,B: HP group,C: normal rats as the control group.Plasma liver functional examination.Liver tissue samples were biopsied for histological study by optic-microscopy.Twelve rats both A and B group were left to observe one week survival rate. Results: The functional lesion of the liver was lessened and cell morphology lesions were slightly compared group B with group A.The one week survival rate in group B was significantly longer than that in group A(P<0.05). Conclusion: HP may lessen the pathological lesions of the liver and improve the one week survival rate by elevating hepatocyte tolerance to serious hypoxia damage during orthotopic liver transplantation.
