长期运动对大鼠十二指肠一氧化氮合酶表达的调节作用

摘    要:目的:观察长期运动及应用一氧化氮合酶抑制剂(L-NAME)对大鼠十二指肠一氧化氮合酶(NOS)表达的调节作用.方法:健康雌性SD大鼠,随机分为四组,即静息组、静息并应用L-NAME组、运动组、运动并应用L-NAME组,L-NAME组的大鼠饮用水中含有L-NAME 1 g/L,后两组大鼠游泳3个月,应用免疫组织化学技术分析大鼠十二指肠神经型NOS(nNOS)和诱导型NOS(iNOS)的表达.结果:与静息组相比,运动组大鼠十二指肠nNOS及iNOS活性均明显加强,前者主要发生于绒毛上皮,后者主要发生于腺上皮和肌层.静息并应用L-NAME组的nNOS阳性表达强于静息组,而两组间iNOS阳性表达无统计学意义.运动并应用L-NAME组绒毛上皮的nNOS阳性表达和腺上皮的iNOS阳性表达均弱于运动组.结论:运动可增强十二指肠黏膜不同部位和不同类型NOS的表达,即运动可增强绒毛上皮的nNOS表达和十二指肠腺上皮iNOS的表达,推测前者可能介导肠道吸收功能的调节,后者可能参与了黏膜分泌功能的调节.

Abstract:Objective: To observe the regulatory role of long-term exercise in the expression of duodenal nitric oxide synthase(NOS).Methods: Sprague-Dawley rats were divided into four groups: a sedentary group,a sedentary plus L-NAME group,an exercise group and an exercise plus L-NAME group.L-NAME was administrated into the L-NAME groups by their drinking water(1 g/L).The rats in exercise group swam for three months,and then,the duodenum nNOS and iNOS expression was analyzed by immunohistochemistry. Results: As compared the sedentary rats,the exercised rats had the increased duodenal expression of nNOS and iNOS,the former markedly expressed in the epithelial cell of duodenal mucosal villus,and the latter mainly expressed in the duodenal gland and smooth muscle.The sedentary plus L-NAME group had the increased nNOS expression and the similar iNOS expression than the sedentary group.Either nNOS in the duodenal mucosal villus or iNOS in the duodenal gland in the exercise plus L-NAME group was weakened as compared to that in the exercise group. Conclusion: Exercise could increase the NOS expression in the different regions of the duodenum mucosal.Exercise may enhance the expression of the nNOS isoform in the duodenal mucosal villus and the iNOS isoform in the duodenal gland.The results suggest that the former may regulate the duodenal absorption and the latter may regulate the mucosal secretion.
