摘    要:目的:探讨4种Notch受体基因在类风湿性关节炎(RA)患者外周血单个核中的表达及其意义.方法:采用免疫磁珠分离纯化RA患者外周血T,B及单核细胞,定量PCR检测4种Notch受体mRNA表达水平,并与正常人进行比较.结果:与正常人相比,RA患者T细胞Notch 2,Notch 3及Notch 4表达量增加(P<0.01),其中Notch 3主要表达于活化的T细胞,而Notch 1水平在两者间未见明显差异.B细胞及单核细胞4种Notch受体表达均较低,且两者间亦未见显著差异.结论:RA患者存在Notch基因表达的改变,一群高表达Notch 3基因的活化T细胞可能在RA疾病机制中具有重要的意义.

Abstract:Objective: To explore the mRNA expression and potential significance of Notch receptors in peripheral blood mononuclear cells.Methods: Peripheral blood T cells,B cells and monocytes were purified by using Dynal immunobeads and Notch mRNA expression was analysed by real-time PCR. Results: Compared with healthy control,T cells from RA patients expressed higher level of Notch 2,Notch 3 and Notch 4,and Notch 3 was mainly expressed by activated T cells,while no difference of Notch 1 expression in T cells or four Notch receptors expression in B cells and monocytes was observed between RA patients and healthy controls. Conclusion: In RA patients,Notch mRNA expression level was changed and a T cell group expressing high level Notch 3 may play an important role in the mechanism of RA.
