基质金属蛋白酶-2及其组织抑制因子表达失衡与卵巢上皮性肿瘤血管生成的关系

摘    要:目的探讨基质金属蛋白酶-2(MMP-2)和基质金属蛋白酶组织抑制物-2(TIMP-2)在卵巢上皮性肿瘤血管生成中的作用及临床意义。方法应用免疫组化SABC法检测20例卵巢良性上皮性肿瘤组织、20例卵巢交界性上皮性肿瘤组织、50例卵巢上皮性癌组织中MMP-2及TIMP-2的表达及微血管密度(MVD)。结果MMP-2阳性表达组的MVD值明显高于阴性表达组的MVD值(P=0.000),卵巢肿瘤组织中MMP-2的表达水平与TIMP-2的表达呈负相关。结论MMP-2和TIMP-2表达失衡可能在卵巢上皮性肿瘤血管生成中起重要作用,检测MMP-2和TIMP-2的表达可作为反映卵巢癌侵袭和转移潜能的生物学指标。

Abstract:Objective To investigate the expression and clinical significance of matrix metalloproteinase-2(MMP-2) and tissue inhibitor of matrix metalloproteinase-2 (TiMP-2) and their correlations with angiogenesis in epithelial ovarian neoplasm.Methods SABC immunohistochemistry was used to determine the expression of MMP-2,TIMP-2 and microvessel density (MVD) in 90 cases of epithelial ovarian neoplasm.Results The MVD value in MMP-2-positive group was much higher than that in MMP-2-negative group(p=0.000).There was a negative correlation between the intensity of the expression of MMP-2 and that of TIMP-2 in epithelial ovarian neoplasm.Conclusion In epithelial ovarian neoplasm,unbalanced expression of MMP-2 and TIMP-2 may play an important role in tumor angiogenesis.MMP-2 and TIMP-2 may be regarded as a biomarker for the potentialities of invasion and metastasis of epithelial ovarian carcinoma.
