摘    要:目的探讨iNOS基因在肿瘤过程中对P53基因的表达和肿瘤细胞增殖活性的影响。方法采用了RNAi技术,将iNOS基因的特异性短发夹状RNA(short hairpin RNA,shRNA)片断转染入舌鳞癌细胞Tca113中,qRT-PCR检测iNOS和P53基因的表达改变情况;CCK8法检测Tca8113细胞的增殖变化情况。结果 RNA干扰iNOS基因后,iNOS基因的表达得到明显降低;而P53基因的表达则有所增加。转染后实验组和对照组间差异有统计学意义(PqPCR<0.05)。CCK8法测定实验组细胞的增殖活性明显低于对照组(PCCK<0.05)。结论通过RNAi降低iNOS基因的表达,可以提高Tca8113细胞中P53基因的表达水平;并降低Tca8113细胞的细胞增殖活性。

Study on expression of P53 and the influence on proliferation activity of Tca8113 cells via RNAi iNOS

Abstract:Objective To research the expression of P53 and the proliferative activity of Tca8113 cells via RNAi iNOS.Methods By RNAi technology,the short hairpin RNA(shRNA) of iNOS was transfected to Tca8113 cells,the change of iNOS and P53 gene expression was detected and the proliferative activity of Tca8113 cells was assayed by CCK8 experiment.Results The expression of iNOS gene significantly decreased,while that of P53 gene was increased after RNA interference iNOS gene.There was statistically significant difference between the experimental and control groups(PqPCR<0.05) after transfection.The proliferative activity of the cells of the experimental group was significantly lower than that of the control group from the result of CCK8(PCCK<0.05).Conclusion Down-expression of iNOS gene by RNAi can raise the level of expression of P53 and reduce the proliferation activity of Tca8113 cells.
