摘    要:目的用ConA通过尾静脉注射、腹腔注射两种给药途径对C57小鼠分别建立急、慢性肝损伤模型,观察ConA不同给药途径造模是否存在差异,并研究ConA造成肝损伤的作用机制。方法用6mg/kg的ConA分别通过尾静脉注射、腹腔注射方法干预C57小鼠,注射8h后处死小鼠建立急性肝损伤模型;用6mg/kg的ConA每周注射1次,连续4周,建立慢性肝损伤模型,观察两种给药途径能否成功建立慢性肝损伤模型,及造成的慢性肝损伤程度有无差异。结果在ConA用量为6mg/kg时通过尾静脉注射能成功的建立急、慢肝损伤模型。而用腹腔注射给药建立急、慢性肝损伤模型存在结果不稳定性,不能成功建立肝损伤模型。结论给药途径对于ConA的药效发挥存在影响,尾静脉注射造成的肝损伤更为严重。

Comparison of Acute and Chronic Hepatic Injury Models in C57 Mice Caused by ConA from Different Routes of Administration

Abstract:Objective With ConA through tail vein injection, intraperitoneal injection of two kinds of route of administration on C57 mice were established acute, chronic liver injury model, obserw; the ConA of different routes of administration of the mold whether differences exist, and to study the mechanism of hepatic injury caused by ConA. Methods 6mg/kg ConA were injected through the tail vein injection of C57 mice, intraperitoneal injection of 8h intervention, mice were sacrificed after establishment of acute liver injury model; use 6mg/kg ConA for once -weekly injection, continuous around, establishment cf chronic liver injury model observe whether two routes of administration can be successfully established the model of chronic liver injury and the chronic liver injury degree has difference. Result Acute or chronic hepatic injury models can be successfully established in mice through tail vein injection with ConA of 6 mg/kg, while those established by intraperitoneal injection are not stable, which can notsuccessfully set up hepatic injury models. Conclusion Drug administration had great effects on ConA. Liver injury in tail vein injection caused more serious; than intraperitoneal injection.
