透骨消痛胶囊干预膝骨性关节炎软骨下骨重塑分子机制探讨

摘    要:目的 观察透骨消痛胶囊干预骨性关节炎软骨下骨重塑的作用及机制.方法 128只新西兰大白兔随机分4组,除正常组外均按改良Hulth法造模.正常组、模型组予生理盐水10 mL/d灌胃,对照组予壮骨关节丸水溶液10 mL/d灌胃,实验组予透骨消痛胶囊水溶液10 mL/d灌胃,术后分批分次处死取材.RT-PCR法检测软骨下骨c-fos、β-Catenin、RANKL和软骨MMP-1 mRNA的表达.结果 与正常组比较,造模后6周、9周、12周,实验组各指标表达均较高(P<0.05).与模型组比较,造模后1周,实验组c-fos mKNA表达较低(P<0.05);造模后6周,实验组RANKL、MMP-1 mRNA表达较低(P<0.05);造模后9周和12周,实验组各项指标表达均较低(P<0.05).与对照组比较,造模1周后,实验组c-fos mRNA表达较低(P<0.05);6周时实验组RANKL、MMP-1 mRNA 表达较低(P<0.05);9周、12周后,实验组β-Catenin、MMP-1 mRNA表达较低(P<0.05).结论 透骨消痛胶囊可改变软骨下骨重塑的速率和模式,最终减轻软骨下骨硬化.
