BrdU标记的骨髓来源间充质干细胞在心脏移植部位数量的变化

目的 :观察BrdU对骨髓来源间充质干细胞的标记率及移植后的骨髓来源间充质干细胞在心脏移植部位不同时间中的数量变化情况。方法 :酶标仪分别测出骨髓来源间充质干细胞及BrdU阳性间充质干细胞光密度值 ,计算标记率。将经BrdU标记后的骨髓来源间充质干细胞注射入冠状动脉左前降支中段结扎处的大鼠左心室壁内 ,术后第 5、10、15、2 0天行抗BrdU抗体免疫组织化学染色。结果 :BrdU对骨髓来源间充质干细胞的标记率约为 4 1.4 1%。移植后第 5天BrdU阳性细胞数多于移植后第 10天 (P <0 .0 5 ) ,而移植后第15、2 0天与第 10天相比BrdU阳性细胞数无明显变化 (P >0 .0 5 )。结论 :BrdU能对骨髓来源间充质干细胞起到较好的标记和“跟踪”作用 ,移植入心脏壁内的间充质干细胞数量第 10天比第 5天减少 ,以后基本稳定的变化过程。

The Variations of BrdU-labeled Marrow-derived Stromal Cells after Transplanted into Heart Mural

Objective:To observe BrdU labeling efficiency of marrow-derived stromal cells and the variations after transplanted into heart mural. Methods: By means of scanning multiwell spectrophotometer, optical density of marrow-derived stromal cells and BrdU-positive stromal cells was measured respectively. The labeled marrow-derived stromal cells were injected into the left ventricular wall where the left anterior descending branch were ligated, and then immunohistochemical staining was done at 5-, 10-, 15-, 20-day intervals after the cells implantation surgery. Results: BrdU labeling efficiency of marrow-derived stem cells was 41.41%. The experiment showed that BrdU-positive cells on the 10th, 15th and 20th day were less than that on the 5th day(P<0.05), but there existed no significant variance among the 10th,15th,and 20th day(P>0.05). Conclusion: BrdU can label marrow-derived stromal cells effectively and the transplanted cells decreased in early period.
