盐酸小檗碱对血管平滑肌细胞增殖的干预研究

摘    要:目的:探讨盐酸小檗碱对抑制血管平滑肌细胞增殖的干预机制。方法:分离培养大鼠血管平滑肌细胞(vascularsmoothmllsclecells,VSMC),并分为实验组,对照组及空白组;MTT法选择盐酸小檗碱干预浓度;MTT法和3H]-胸腺嘧啶核苷渗入法检测VSMC增殖率;Westernblotting方法检测ERKl/2、Akt的磷酸化蛋白的变化。结果:VSMC实验组细胞增殖率与对照组相比明显下调,P〈0.05;小檗碱干预后,VSMC细胞ERKl/2、Akt的磷酸化蛋白的表达明显下调,与对照组相比,P〈0.05。结论:盐酸小檗碱对血管平滑肌细胞的增殖有一定的抑制作用,可能与其抑制ERKl/2、Akt等的表达有关。

Abstract:Objective:To investigate the mechanisms regarding berberine attenuates vascular smooth muscle cell proliferation. Methods: Vascular smooth muscle cell (VSMC) in rats were Isolated, cultured and allocated to experimental group, control group and blank group. The concentration of berberine was determined by using the thiazolyl blue (MT]') approach, and the rate of VSMC proliferation was assayed by MTF and 3H ]-thymidine infiltration method. Western blotting was employed to detect ERK1/2 and Akt expression. Results: Compared with control group, the proliferation of VSMC in experimental group was suppressed by berberine ( P 〈 0.05 ). Intervention with berberine resulted in marked down-regulated phosphorylated proteins of ERK1/2 and Akt (P 〈 0.05 ). Conclusion : The inhibitory effects of berberine on VSMC proliferation may be related to suppression of ERK1/2 and Akt expression.
