咳喘宁雾化吸入对哮喘大鼠肺泡灌洗液白介素-5与嗜酸性粒细胞的影响

摘    要:目的 观察咳喘宁雾化吸入对肺泡灌洗液(BALF)中白介素-5(IL-5)表达、嗜酸性粒细胞(EOS)计数的影响,探讨其对哮喘大鼠气道炎症的作用机制.方法 将SD雄性幼龄大鼠40只,随机分为正常组、哮喘模型组、咳喘宁干预组、布地奈德干预组,每组10只.经鸡卵蛋白(OVA)致敏后,给予OVA雾化吸入诱发哮喘发作,制作哮喘模型.咳喘宁干预组与布地奈德干预组分别给予咳喘宁口服液1.7 g/ks,布地奈德0.1 mg/kg,正常组与哮喘模型组予以等量生理盐水雾化.雾化7d后,处死大鼠,检测大鼠BALF中IL-5水平和EOS计数.结果 咳喘宁干预组大鼠BALF中的IL-5与EOS明显低于哮喘模型组,差异有统计学意义(P<0.05),与布地奈德干预组比较,差异无统计学意义(P>0.05).结论 咳喘宁通过雾化吸入能降低哮喘大鼠BALF的IL-5水平及EOS计数,从而改善哮喘大鼠气道炎症状态.

Abstract:Objective To observe the effects of Kechuanning(KCN) by atomization inhalation on the expression of interleukin 5(IL-5) and eosinophil(EOS) count in bronchoalveolar lavage fluid (BALF) in asthmatic SD rats,and explore the mechanism of KCN on airway inflammation of asthmatic SD rats.Methods 40 SD rats were randomly divided into normal group,asthmatic group,KCN group and budesonide group with 10 rats in each group.The asthmatic model was established by sensitizing and challenging SD rats with aerosolized ovalbumin (OVA).KCN group rats were treated with 1.7 g/kg KCN by atomization inhalation,budesonide group rats were treated with 0.1 mg/kg budesonide,normal group and asthmatic group rats were treated with equal amount of saline.After treatment for a week,the rats were killed and the IL-5 in BALF was measured by enzyme linked immunosorbent assay,EOS count was also carried out.Results The IL-5 and EOS of BALF in the KCN group rats were markedly lower than those in asthmatic group (P<0.05).The differences between the KCN group and the budesonide group were not marked (P>0.05).Conclusion KCN by atomization inhalation can decrease the level of IL-5 and EOS in BALF,and then improve airway inflammation of asthmatic rats.
