活血利水法对外伤性实验性PVR模型兔眼玻璃体中生长因子浓度影响的研究

摘    要:目的探讨活血利水之散血明目片对外伤性增生性玻璃体视网膜病变 (traumaticProliferativeVit reoretinopathy,tPVR)玻璃体中生长因子浓度的影响及防治PVR的作用机理。方法将 5 0只家兔随机抽取 40只 ,造成外伤性PVR模型 ,随机分成活血利水组、活血化瘀组、利水明目组、模型组 ,另 1 0只为空白组。连续灌胃 1月后 ,观察眼底PVR分级情况 ,检测玻璃体中碱性成纤维细胞生长因子 (bFGF)、表皮生长因子 (EGF)的含量及病理组织学改变。结果活血利水组中玻璃体碱性纤维细胞生长因子 (bFGF)、表皮生长因子(EGF)、浓度低于模型组 ,差异有非常显著性 (P <0 .0 1 ) ;活血利水治疗组EGF含量与空白组比较差异无显著性 (P >0 .0 5 ) ,bFGF含量高于空白组 (P <0 .0 5 ) ;活血化瘀组与利水明目组也能降低bFGF及EGF浓度 ,与模型组相比有显著性意义 (P <0 .0 5 ) ;活血利水组bFGF及EGF浓度低于另两个治疗组 ,有显著性差异 (P<0 .0 5 )。结论活血利水法能明显降低玻璃体中碱性成纤维细胞生长因子和表皮生长因子的浓度 ,抑制增殖细胞的过度增生从而防治PVR形成和发展

Abstract:Objective To investigate the effects of blood-quickening water-disinhibiting method (BQWD) on the contents of growth factors in vitreous body of modeling rabbit eyes with experimently induced traumatic proliferative vitreoretinopathy (tPVR).Methods Fourty rabbits were prepared as tPVR model at first.Then, they were randomly divided into 4 groups, i.e.group 1 (G1) treated with BQWD herbs,group 2 (G2) treated with blood-quickening stasis-transforming (BQST) herbs,group 3 (G3) treated with water-disinhibiting eye-brightening (WDEB) herbs and grous 4 (G4) treated with no herbs.Another 10 healthy rabbits were taken as blank group (G5).The treatment was lasted for one month by forced perfusing gastrically. Thereafter, the indexes to be detected included PVR grading,histopathological changing and the contents of basic fibroblast growth factor (bFGF) and epidermal growth factor (EGF) in vitreous body.Results When campared with that of modeling group (G4),bFGF and EGF contents were both significantly lower in BQWD group (G1,P<0.01),and such changes were also true in BQST group (G2) and WDEB group (G3),P<0.05.When compared with that of blank group (G5),bFGF content was significantly higher in G1(P<0.01),while there was no significant difference present in the contents of EGF between them.Furthermore, there were also significant differences in the contents of bFGF and EGF among G1 and the other two treating groups (G2 and G3),with obviously lower values in BQWD group (P<0.05).Conclusions BQWD method can not only treat tPVR but also prevent it from developing via the mechanism of declining the contents of bFGF and EGF in vitreous body to inhibit hyperplasia lesion taking place in ocular fundus.
