痛风速效宁对大鼠急性痛风性关节炎的影响

目的研究痛风速效宁对高尿酸血症并急性痛风性关节炎模型大鼠不同时相的影响,探讨其治疗急性痛风性关节炎的作用机制。方法将雄性SD大鼠96只随机分为3组,模型对照组(A)、秋水仙碱组(B)、痛风速效宁组(C),用氧嗪酸钾和尿酸钠分别行腹腔和踝关节腔内注射制造高尿酸血症并急性痛风性关节炎模型,各组于造模前3 h和造模后3 h分别予灌胃生理盐水、秋水仙碱和痛风速效宁干预,并于造模后6、12、24、36 h 4个时相各取8只大鼠,采血、取关节液和滑膜,比较3组在血尿酸、关节液白细胞和尿酸盐晶体方面的变化。结果①痛风速效宁可降低实验大鼠的血尿酸水平,与模型对照组比较P<0.01;②痛风速效宁可加速致炎大鼠的关节液中尿酸盐晶体的清除,与模型对照组比较P<0.01;③痛风速效宁可抑制致炎大鼠的关节液中WBC渗出、抑制和改善致炎大鼠的关节滑膜炎症反应程度,与秋水仙碱组比较P>0.05,与模型对照组比较P<0.05。结论痛风速效宁对高尿酸血症并急性痛风性关节炎模型大鼠具有治疗作用。

Experimental Study of Tongfeng Suxiaoning on Acute Gouty Arthritis in Rats

Objective To observe the effect of Tongfeng Suxiaoning(TFSXN) on the hyperuricemia and acute gouty arthritis in different period in rats,and investigate the curative mechanism of TFSXN in the arthritis.Method The 96 SD male rats were randomly divided into three groups as model control group(A),colchicine group(B) and TFSXN group(C),32 rats in each group.The models with hyperuricemia and acute gouty arthritis were induced by uricase inhibitor potassium oxonate injected into abdominal cavity and urate sodium solution into the articular cavity of knee joints;3 hours before and after the establishment of the model,physiological saline and colchicine were filled into intra-gastric in model contrast and colchicines groups,and TFSXN into TFSXN group;Then,the blood,articular fluid and synovium in the joints were obtained from 8 rats in each group in the periods of 6,12,24,36 hours to measure the changes of the serum uric acid,leucocytes in joint fluid and urate sodium crystal.Result Compared with A,1.TFSXN decreased the level of serum uric acid(P<0.01);2.TFSXN promoted the clearing of urate sodium crystal(P<0.01);and compared with C,3.TFSXN inhibited leucocytes effusion in the joint fluid,relieved and improved the severity of synovial inflammation(P>0.05,P<0.05).Conclusion TFSXN has the curative effect for the hyperuricemia and acute gouty arthritis in rats.
