糖尿病胃轻瘫大鼠模型衍变过程相关指标的观察

摘    要:目的 观察糖尿病胃轻瘫(Diabetic Gastroparesis DGP)大鼠模型衍变过程中相关指标的变化.方法 将造模组大鼠按55 mg/kg单次腹腔注射2%链脲佐菌素(STZ)配合高糖高脂饲料不规则喂养8周建立DGP模型,检测血糖、尿糖变化;以酚红为标记物测胃排空率及小肠推进率;每周观察并记录体质量、24h饮水量、食量、尿量;以自制评分表评定一般症状情况.结果 与空白组比较,STZ造模组大鼠血糖、尿糖值显著升高(P<0.01),胃排空率及小肠推进率明显降低(P<0.01);由食量、饮水量、尿量增多,体质量减轻逐渐变为饮水量、尿量增多,体质量、食量减少并伴有精神、行动、皮毛、大便性状明显改变.结论 以2%STZ的55 mg/kg联合高糖高脂不规则饮食成功建立DGP大鼠模型;观察此模型衍变过程的各项相关指标改变的时间点及变化的程度,完善了以往缺乏症状改变情况详细记录的DPG造模过程.

Abstract:Objective To observe the variation of basic indicators in the evolvement process of diabetic gastroparesis (DGP) rat models. Methods According to the result of preliminary experiment. The rats were cultured by single intraperitoneal injection of 2% STZ (55 mg/kg) and combined with high-sugar and high-fat fodder irregularly for 8 weeks to build the DGP models. The blood sugar and urine sugar were monitored; the rate of gastric emptying and small intestinal propulsion rates were measured by phenol red as markers; the body weight, 24-hours drinking volume, urine volume and food intake were recorded every week; the general symptoms were assessed by self-made scale. Results Compared with the blank group, blood sugar and urine sugar of STZ modeled group rats were significantly increased (P〈0.01), the rate of gastric emptying and small intestinal propulsion rates were obviously decreased (P〈0.05 or P〈0.01). By the commonsymptoms which showed the increase of food intake, drinking volume, urine volume and loss of weight gradually become the states that drinking volume and urine volume increased along with weight reduction and the decrease of food intake. Conclusion The combination of 55 mg/kg 2% STZ with high-sugar and high-fat irregular diets could induce DGP rat models successfully. The observation of models evolvement processes would reflect the transformation time and the level of various basic indicators, and then improve the process of DPG models lacked of previous detailed records of symptom changes.
