补阳还五汤对局灶性脑缺血大鼠脑内caveolin1、2的影响

摘    要:目的探讨补阳还五汤对局灶性脑缺血大鼠脑内caveolin1、2的影响。方法采用大脑中动脉阻塞法复制脑缺血模型,将动物随机分为正常组、假手术组、模型组、补阳还五汤组,分别给予不同处理,在12 h、1、3、7 d不同时间点处死动物,采用免疫组织化学法观察caveolin1、2的表达。结果补阳还五汤能显著减轻神经功能缺损;正常脑内有一定量的caveolin1、2表达,缺血后caveolin1、2迅速增加,分别于术后3 d或1 d达高峰,补阳还五汤组显著提高caveolin1、2的表达,在3、7 d时模型组与补阳还五汤组差异有统计学意义(P<0.01)。结论补阳还五汤可调节脑缺血后caveolin1、2的表达,是其促进神经功能恢复的可能机制之一。

Abstract:Objective To investigate the effects of Buyang Huanwu decoction(BYHW) on expression of caveolin1 and 2 of cerebral ischemia in rats.Methods Cerebral ischemia was induced by middle cerebral artery occlusion.The rats were randomly divided into four groups including naive,shame-operated,control and BYHW groups.All the rats were killed in 12 hours and 1,3 or 7day after operation.The expressions of caveolin1 and caveolin2 were measured by immunohistochemical staining.Results BYHW could reduce obviously the injury of nervous functions;expression of caveolin1 and 2 positive cells existed in normal brain,caveolin 1 or 2 were enhanced immediately after ischemia and reached to peak at 3 day or 1 day after ischemia respectively;BYHW could increase the number of caveolin1 and 2 positive cells at 3 and 7 day after ischemia and treatment(P<0.01).Conclusion BYHW is capable of regulating expression of caveolin1 and 2 in ischemic brain which may be one of anti-ischemic mechanism of the decoction.
