尼莫地平对糖尿病大鼠海马突触素蛋白和mRNA表达的影响

摘    要:目的:观察尼莫地平对糖尿病大鼠海马突触素表达的影响.方法:将38只Wistar大鼠随机分为正常对照组、糖尿病组、糖尿病 尼莫地平组.建立STZ糖尿病大鼠模型,尼莫地平干预12周,Morris水迷宫测试其学习记忆能力,免疫组化观察大鼠海马突触素蛋白表达,RT-PCR检测突触素mRNA水平表达变化.结果:尼莫地平组与糖尿病组比较,Morris水迷宫测试中潜伏期明显缩短(P<0.05),中心区停留时间百分比和通过原平台位置次数增加(P<0.05),突触素蛋白和mRNA表达均增加(P<0.05).结论:尼莫地平对糖尿病大鼠认知功能的保护作用可能与其使海马突触素表达增加有关.

Abstract:Objective:To explore the effect of Nimodipine on synaptophysin in diabetic rat.Methods:38 Wistar rats were divided into the normal control group, diabetic model group, the Nimodipine treatment group randomly. Streptozotocin-diabetic rat model was established. The diabetic rats were treated with Nimodipine for 12 weeks. Then, the learning and memory abilities of rats were tested with the Morris water maze.The mRNA and protein level of synaptophysin in hippocampus were detected by RT-PCR and immunohistochemical method. Results:Compared with diabetic model group, the Nimodipine treatment group showed a significant decrease in the mean time of escape latencies(P<0.05)and a increase in percentage of stay time in the the central area and the number of passing the original platform position(P<0.05).And the expression of synaptic protein and mRNA in hippocampus were elevated significantly(P<0.05).Conclusion:It is concluded that Nimodipine could improve cognitive ability in diabetic rat through increasing the expression of synaptophysin.
