db/db自发性糖尿病小鼠颌下腺EGFR的表达及细胞凋亡关系的研究

摘    要:目的:观察db/db自发性糖尿病小鼠颌下腺中表皮生长因子受体(EGFR)的表达及其与细胞凋亡的关系. 方法: 选取3,4,6,8,10月龄db/db糖尿病小鼠及相应月龄的db/ m正常小鼠颌下腺,应用SP免疫组织化学方法染色及TUNEL原位标记后进行图像分析,统计EGFR及凋亡细胞在颌下腺组织内表达(分布)的细胞阳性率. 结果:EGFR及凋亡细胞在对照组及糖尿病组颌下腺中均有表达(分布). db/db糖尿病小鼠颌下腺EGFR及凋亡细胞阳性率随病程延长均呈增高趋势,且显著高于对照组. 结论:EGFR表达增多,说明糖尿病时EGFR作为多效应受体,被激活后可能诱导了颌下腺的细胞凋亡. 凋亡细胞阳性率的显著增高,提示糖尿病是加剧颌下腺腺体萎缩,功能受损的原因.

Abstract:AIM: To observe the expression of EGFR and the distribution of apoptotic cells in submandibular gland of db/db mutant diabetic mice and to explore their relationship. METHODS: Submandibular glands were harvested from the selected 3, 4, 6, 8, 10 month-old db/db diabetic mice (n=25) and the age-matched db/+m normal mice (n=25); Every age subgroup included 5 mice. The expression of EGFR and the distribution of apoptotic cells were observed by using immunohistochemistry and TUNEL.The results were analyzed statistically. RESULTS: EGFR was expressed and apoptotic cells were pre-sented in submandibular gland from both diabetic mice and control mice. The percentage of EGFR-immunopositive cells and the number of apoptotic cells were significantly higher in diabetic mice than those of control mice, and they showed increasing tendency as disease progressed in diabetic groups. CONCLUSION: EGFR-immunopositive cells increase with the development of the disease, which may lead to the apoptosis of epithelial cells. The increase of apoptotic cells demonstrates that diabetes mellitus can induce apoptosis, which may be the reason of submandibular gland atrophy.
